摘 要:目的:观察和探讨灵芝孢子粉对戊四氮(PTZ)致痫大鼠海马区肿瘤坏死因子-α(TNF-α)、内皮素-1(ET-1)含量变化的影响。方法:用亚惊厥剂量的戊四氮(PTZ)复制慢性癫痫模型,用药组以灵芝孢子粉灌胃,记录癫痫发作的级别及潜伏期,并采用放射免疫法检测各组海马区TNF-α、ET-1的含量。结果:癫痫模型组与正常对照组相比,TNF-α、ET-1的含量明显增高(P<0.01);灵芝孢子粉组与癫痫模型组相比,造模第17、21、25天时癫痫发作的潜伏期二者差异有显著性(分别为P<0.05;P<0.05;P<0.01),TNF-α、ET-1的含量明显降低(P<0.05)。结论:TNF-α、ET-1可能参与了PTZ致痫的发生发展过程,灵芝孢子粉能明显减弱TNF-α、ET-1过度表达对癫痫大鼠中枢神经系统的毒性损害,可能是通过调节神经系统免疫炎症反应而实现的,对癫痫发作导致的神经细胞损伤具有保护作用。Objective:To observe and explore the influence of Ganoderma lucidum spores on the TNF- α and ET-1 content in the hippocampus of rat with epilepsy caused by Pentetrazole(PTZ). Methods: We used a sub-eclampsia dose of PTZ to develop epilepsy model, gave Ganoderma lucidum spores into the rat stomach in the Ganoderma lucidum spores group, recorded the spasm level and latent period, and then measured the content change of TNF-α and ET-1 in the hippocampus. Results:Compared with contrast group, the TNF-α and ET--1 content in epilepsy model group increased obviously (P 〈0.01 ); the latent period of Ganoderma lucidum spores group showed obvious difference at 17, 21 and 25 days compared with epilepsy model group ( P 〈 0.05;P 〈 0.05;P 〈 0.01 , respectively ). Conelusion:TNF-α and ET-1 may be involved in the occurrence and development of PTZ-caused epilepsy, and Ganoderma lucidum spores can weaken the noxious injury caused by over expression of TNF-α and ET-1 in the rat centre nerve system,and therefore, prevent the damages to nerve cells by epilepsy onset.
