摘 要:目的:观察Ⅰ,Ⅲ型前胶原核酶对裸鼠增生性瘢痕的作用效果。方法:实验于2004-02/06在中山大学医学院实验动物中心、电子显微镜室及第一附属医院外科实验室进行。①建立裸鼠增生性瘢痕动物模型(80只)。②将裸鼠随机分为实验组Ⅰ型组20只,注射Ⅰ型前胶原核酶;Ⅲ型组20只,注射Ⅲ型前胶原核酶;Ⅰ型+Ⅲ型组20只,注射Ⅰ型及Ⅲ型前胶原核酶;各组又分为2.5μg/100μL及5.0μg/100μL两个剂量组,每个剂量组各10只裸鼠。对照组:生理盐水组10只,注射生理盐水;空白对照组10只。于建立模型的第4周开始,实验组与生理盐水组每天用微量注射器行瘢痕内多位点注射1次,共7d;第6周取材。③应用苦味酸-天狼猩红染色偏振光法,结合图像分析技术,观察分析各组胶原含量及分布的改变。④电子显微镜观察各组实验前后超微结构的改变。结果:①各实验组瘢痕体积均缩小;Ⅰ型组、Ⅰ型+Ⅲ型组实验后与实验前比较体积明显缩小[Ⅰ型组2.5μg/100μL:(25.6±1.2),(28.3±2.1)mm3,5.0μg/100μL:(27.9±3.0),(30.4±1.7)mm3;Ⅰ型+Ⅲ型组2.5μg/100μL:(24.8±1.6),(29.5±3.3)mm3,5.0μg/100μL:(24.6±1.7),(27.8±1.8)mm3,t=1.981~2.025,P<0.05]。②Ⅰ型组、Ⅰ型+Ⅲ型组与对照组相比Ⅰ型胶原纤维分布明显减少。③生理盐水、空白对照组具有典型的增生性瘢痕的特点,各实验组瘢痕形态改变不明显。④实验组及对照组的超微结构改变无明显差异。结论:Ⅰ和Ⅲ型前胶原核酶均可有效地减少裸鼠增生性瘢痕中胶原的含量。AIM: To study the effect of ribozymes of type Ⅰ and Ⅲ procollagen on hyperplastic scar of nude mice. METHODS: The experiment was conducted at the experimental animal center, Medical College, Sun Yat-sen University, Electron Microscope Room and Surgery Laboratory, First At/iliated Hospital from February to June 2004. ① Models with hyperplastic scar in nude mice were established (80 mice). ② The nude mice were assigned randomly into experimental group: type Ⅰ group with 20 mice, injected with ribozymes of type Ⅰ procollagen; type Ⅲ group with 20 mice, injected with ribozymes of type Ⅲ procollagen; type Ⅰ and Ⅲ group with 20 mice, injected with ribozymes of type Ⅰ and Ⅲ of procollagen, and each group was divided into 2.5 μg/100 μL and 5.0 μg/100 μL dosages groups with 10 nude mice in every group. Control group: saline group with 10 mice, injected with saline; blank control group with 10 mice. From the 4^th week after building model, injection was performed into multiple sites with microsyringe in the experimental group and saline group, once every day for 7 days. Materials were gained at the 6^th week. ③ The changes of content and distribution of collagen in every group were observed and analyzed with carbazotic acid-sirius red stain and polarization microscopy, combined with image analysis. ④ Changes of uhrastructure were observed before and after experiment in every group with electron microscope. RESULTS: ① The scar volume was reduced in experimental groups. The -volume shortened in the type Ⅰ and Ⅰ + Ⅲ groups after experiment than that before experiment [type Ⅰ group 2.5 μg/100 μL: (25.6±1.2), (28.3 ±2.1) mm^3, 5.0 μg/100 μL: (27.9±3.0), (30.4±1.7) mm^3; type Ⅰ + Ⅲ group 2.5 μg/100 μL : (24.8±1.6), (29.5±3.3) mm^3, 5.0 μg/100 μL: (24.6±1.7), (27.8±1.8) mm^3,t=1.981-2.025, P 〈 0.05]. ② The distribution of type Ⅰ collagenous fiber diminished obviously in the type Ⅰ group and type Ⅰ
