摘 要:目的:探讨三七皂苷Rg1对大鼠局灶性脑缺血再灌注损伤小脑和延髓的BDNF阳性蛋白的含量和阳性神经元数是否具有上调作用.方法:选取成年雄性SD大鼠60只,随机分为脑缺血再灌注模型组、药物(高、中、低剂量)干预组和阳性对照组,采用线栓法制作大鼠局灶性脑缺血再灌注模型,各组分别于给药后1、3、7 d随机取4只大鼠处死取材,冰冻法切片,免疫组化ABC法染色,用高清晰度彩色病理图文报告分析系统测量分析各组大鼠小脑和延髓的BDNF阳性蛋白的含量和阳性神经元数目的变化,并观察大鼠脑缺血后的神经缺失症状,数据用方差分析和q检验进行统计学处理.结果:与模型组相比,三七皂苷Rg1高、中、低剂量组在给药1、3、7 d后均能明显改善脑缺血的神经缺失症状,并能上调大鼠脑缺血再灌注损伤小脑和延髓的BDNF阳性蛋白的含量和阳性神经元数量(P<0.05);与阳性对照组相比,在不同组别中,作用上强于或类似于阳性对照药尼莫地平.结论:三七皂苷Rg1能上调BDNF阳性蛋白的表达,通过BDNF对脑缺血再灌注神经元损伤所起的保护作用,从而发挥其对脑缺血的治疗作用,这可能是三七皂苷Rg1对脑缺血保护作用的机制之一.Objective: To study the effects of notoginsenoside - Rg1 on BDNF protein in rat cerebella and medulla oblongata of MCAO/R injury, as well as to investigate whether notogisenoside- Rg1 can up- regulate the protein content of BDNF of the positive neurons and the amount of the positive neurons. Methods. 60 adult male SD rats, weighing 280- 320 g, were randomly divided into model group (0.9 % normal saline treated with 5 mL/kg) ; high dose group ( notoginsenoside - Rg1 treated with 200 mg/kg) ; middle dote group (notoginsenoside-Rg1 treated with 100 mg/kg) and low dose group (notoginsenoside- Rg1 treated with 50,g/kg) ; and the positive control group (nimodipine treated with 1 mg/kg). All drugs were given once a day with ip till the mouse was sacrificed. The focal cerebral ischemia - reperfusion model was made with thread - occluded method. Their frozen brain tissue were sliced into sections of 12μm thickness. The four rats were randomly taken from each group to be treated as specimens after surgical handle in 1, 3, 7 days, respectively. The slices were processed with immunohistochemical ABC techniques. The protein content of BDNF of the positive neurons and the amount of the positive neurons in cerebella and medulla oblongata in each rat were observed and counted by the HPIAS - 1000 analytic system, and the nervous deficit symptoms were observed after the cerebral ischemia. The data were analysed by one - way Anova and q test. Results. Compared with the model group, notoginsenoside - Rg1 could obviously improve .some nervous deficit symptoms after rats' fo- cal cerebral ischemia reperfusion injury. They also could increase the protein content of BDNF of the positive neurons and the amount of the positive neurons in the cerebella and medulla oblongata of focal cerebral ischemia reperfusion injury in rats (P 〈0.05). Compared with the positive control group, the effect of noto- ginsenoside- Rg1 was superior to nimodipine or similar to it. Conclusion: Notoginsenoside - Rg1 could up - reg
