摘  要:目的:探讨来源于不同组织肺泡、腹腔巨噬细胞甘露糖受体(macrophagemannosereceptor,MMR)与配体的亲和力差异。方法:采用支气管肺泡灌洗和腹腔冲洗法获得肺泡和腹腔巨噬细胞(macrophage,Mφ),经纯化、鉴定后与异硫氰酸荧光标记的甘露糖基化牛血清白蛋白(MFITCBSA)孵育20min,分别加入D甘露糖、D半乳糖、EDTA,采用荧光显微镜和流式细胞仪检测甘露糖受体(mannosereceptor,MR)与配体的结合情况。结果:Mφ上存在MR,MR与配体的结合属于Ca2+依赖性,其结合可被D甘露糖、EDTA抑制,而不受D半乳糖的抑制;腹腔MMR与配体的亲和力高于肺泡MMR。结论:腹腔MMR与配体的亲和力高于肺泡MMR。AIM: To evaluate the affinity of the mannose receptor of peritoneal maerophage and puhnonary macrophage and their ligands. METHODS: Two kinds of macrophage were acquired through Irrigation of peritoneal cavity and bronehoalveolar lavage ( BAL). After purification and identification, maerophage were incubated with fluorescent isothioeyanate ( FITC )-labeled mannosvlated albumin (M-FITC-BSA, Sigma A7790) for 20 minites, the affinitv of MR and its ligands are measured by flow cytometry (FCM) and fluoreseene microscope. RESULTS: There existed MR in macrophage. The bindingof MR and its ligands was calcium dependent, which was inhibited by the existence of D-mannose and EDTA, but not by D-galactose. The 'affinity of peritoneal macmphage MR and its ligands were greater than that of the puhnonary macrophage MR and its ligands. CONCLUSION: The afinitv of peritoneal macrophage MR and its ligands are greater than that of the pulmonary macrophage MR and its ligands.
