银屑病患者外周血单一核细胞中趋化因子受体CXCR1及CXCR2 mRNA的表达

摘　　要：目的:探讨银屑病患者外周血单一核细胞(PBMC)中CXC型趋化因子受体CXCR1及CXCR2的表达水平及其临床意义。方法:应用逆转录-聚合酶链反应(RT-PCR)法,检测了进行期斑块状银屑病患者33例及其中16例患者治疗后PBMC中CXCR1及CXCR2 mRNA的表达,设30名健康人作为正常对照,并将检测结果与患者皮损面积及严重程度指数(PASI)进行了相关性分析。结果:斑块状银屑病患者PBMC中CXCR2 mRNA表达水平为1.38±0.87,明显高于健康人对照组0.73±0.58,治疗后患者1.21±0.62仍高于健康对照组(P<0.05);CXCR1 mRNA表达水平为0.26±0.28,与健康对照组及同一组患者治疗后相比均无明显差异,分别为:0.17±0.26,0.27±0.31(均P>0.05);银屑病患者PBMC中CXCR2 mRNA水平高于CXCR1 mRNA水平,二者均与PASI之间无明显相关性。结论:CX-CR2可能参与了银屑病的发病机制。Objective： To investigate the expression of ehemotactic receptors of CXCR1 and CXCR2 in progressive plaque psoriasis and their clinical significance. Methods： Reverse transcription - polymcrase chain reaction （ RT - PCR） technique was applied to detect the expression of CXCR1 and CXCR2 mRNA in peripheral blood mononuclear cells （PBMC） in 33 cases of psoriasis and 30 healthy controls. The expressions of receptors of CXCR1 and CXCR2 after the treatment in 16 cases out of the 33 patients were measured as well. The correlation between CXCR1/2 and psoriatic area and severity index （PAS1） were evaluated. Results： The CXCR2 mRNA level in PBMC in psoriatic patients were 1.38 ± 0.87, which was markedly higher than that in normal controls 0.73 ± 0.58 （ P 〈 0. 001 ） and similar to the treated patients 1.21 ± 0,62. The later were also markedly higher than that in normal controis （P 〈 0.05）. However, there was no statistic significance in CXCR1 mRNA levels in PBMC among untreated psoriatic patients 0.26 ± 0,28, normal controls and the treated patients 0.17 ± 0.26 and 0.27± 0.31, respective- ly. No correlation was noticed both between CXCR1 and PASI and CXCR2 and PASI, Conclusion： The increased expression of CXCR2 may be involved in the pathogenesis of progressive plaque psoriasis.
