摘  要:目的:探讨血管生物反应器中切应力作用下血管内皮细胞(ECs)在PLGA膜片的生长情况.方法:接种血管ECs于可降解基质膜片PLGA上,施加不同切应力,观察细胞生长情况及分泌情况.结果:在培养5~7 d后,ECs生长良好,形态略有伸长,细胞整体上有沿流体方向改变形态的趋势.ECs在PLGA膜片上NO的分泌量为(0.875±0.011)×10-9mol/min.结论:PLGA材料对细胞亲和力较好.ECs释放NO等血管活性物质功能正常.为进一步开展采用生物反应器体外构筑组织工程血管及探索提高组织工程血管内膜层抗血流冲击能力提供实验数据.AIM: To study the growth of vascular endothelial cells(ECs) on PLGA mesh under shear stress in vascular bioreactor.METHODS: After seeded on PLGA mesh,vascular ECs were given shear stress in vascular bioreactor for 7 d and the growth and the externalization of the cells were observed.RESULTS: After cultured for 5-7 d,the vascular ECs grew well and they elongated in appearance.The cells had a tendency to change their appearances(along) the orientation of the fluid.The ECs secretary volume was(0.875±0.011)×10^(-9) mol/min.CONCLUSION: The materials of PLGA have a good affinity to the cells.The ECs' function of releasing NO and other active substances of blood vessels is normal.Primary experimental data have been obtained for further researches on the tissue-engineering vessel construction in vitro by using bioreactors and on the improvement of intima tolerance to blood flow's impact.
