温心胶囊对心力衰竭大鼠心肌基质金属蛋白酶组织抑制物1mRNA表达的干预效应

摘　　要：目的:观察温心胶囊对异丙肾上腺素诱导的充血性心力衰竭大鼠心肌基质金属蛋白酶组织抑制物1mRNA表达的影响,探讨基质金属蛋白酶组织抑制物1在心力衰竭发病中的作用及温心胶囊防治心力衰竭心肌重塑的作用机制。方法:①实验分别于黑龙江中医药大学中医基础理论实验室和哈尔滨医科大学遗传教研室分子生物学实验室完成。②选用健康Wistar雄性大鼠60只。大鼠被随机分为5组,空白对照组,模型组,西药对照组,温心胶囊低、高剂量组,每组12只。③温心胶囊(由桂枝、黄芪、赤芍、茯苓等组成)悬液:每毫升悬液含生药量2g。④除空白对照组外,其余各组皮下多点注射异丙肾上腺素,以每天20,10和5mg/kg的递减剂量连续注射3d,再以每天3mg/kg剂量维持9d。于造模前1天起开始灌胃给药各组连续给药6周。⑤治疗结束后,连接MP100A-CE多导生理记录仪记录左室收缩压、左室舒张末压、室内压最大上升速率、室内压最大下降速率。计算心脏质量指数(心脏湿重/体质量)。进行苦味酸天狼猩红特殊胶原染色法检测,运用彩色病理图像系统分析。计算胶原面积与统计场总面积比值(反映Ⅰ型胶原含量)。应用反转录多聚酶链反应法检测心肌基质金属蛋白酶组织抑制物1mRNA表达。⑥数据间差异性比较采用单因素方差分析。结果:实验过程中模型组,药物对照组,温心胶囊高、低剂量组分别死亡1,2,1,3只,相应组进入心脏质量指数比较的只数分别为11,10,11,9只。①室内压最大上升速率、室内压最大下降速率、左室收缩压:西药对照组和温心胶囊高剂量组明显高于模型组(P<0.05～0.01)。②左室舒张末压:西药对照组和温心胶囊高剂量组明显低于模型组(P<0.05～0.01)。③心脏质量指数:西药对照组和温心胶囊高剂量组明显低于模型组(P<0.05～0.01)。④大鼠心肌I型胶原含量:西药对照组,温心胶囊低AIM： To investigate the effect of Wenxin capsule on tissue inhibitor of metalloproteinases-1 （TIMP-1） expression in congestive heart failure （CHF） rat model induced by isoproterenol, and explore the role of TIMP-1 in CHF and the mechanism of Wenxin capsule in preventing and treating myocardial remodeling. METHOIDS： （1） This experiment was carried out in the Basic Theory Laboratory of Heilongjiang University of Traditional Chinese Medicine and Heredity Laboratory of Harbin Medical University. （2） Sixty healthy male Wistar rats were randomly divided into 5 groups with 12 rats in each group： blank control group, model group, western medicine control group, Wenxin capsule low and high dosage groups. （3）Wenxin capsule （including cassia twig, membranous milkvetch root, red peony root, tuckahoe, etc） suspension： corresponding to 2 g/mL. （4） Except the blank control group, CHF animal models were induced by large dosage subcutaneous injection of isoproterenol with 20, 10 and 5 mg/kg, and then with continuous injection of 3 mg/kg for 9 days. Drugs were given orally from one day before model establishment for 6 weeks. （5After the treatment, MPIOOA- CE multiple physiological recorder was connected to assay left ventricular systolic pressure, left ventricular end-diastolic pressure, the maximal rates of rise of ventricular pressure and the maximal rates of decrease of ventricular pressure were recorded with MP100A-CE multipurpose polygraph. Cardiac index was calculated （cardiac wet mass/body mass）. The special collagen staining method and pathological image analyzer were used to detect type 1 collagen. The expression of TIMP-1 mRNA were detected by reverse transcription-polymerase chain reaction （RT-PCR）. （6） The differences of the data were compared with one-way analysis of variance. RESULTS： There were 1, 2, 1 and 3 rats died in the model group, western medicine control group, Wenxin capsule low and high dosage groups respectively, and finally 11, 10, 11 and 9 rats in the
