摘 要:目的研究当归芍药散(DSS)抑制β-淀粉样蛋白(Aβ)沉积的作用。方法老化组单纯用Aβ1~42液,DSS组同时加入DSS,两组均37℃孵育箱体外孵育7d,运用透射电镜和硫磺素T荧光分析方法观察β-淀粉样蛋白Aβ1~42的聚集和纤维化形成的情况。结果电镜观察显示:老化组其纤维呈晶状聚集,长0.34~1.70μm,直径约10nm。DSS组Aβ聚集不明显,无明显Aβ纤维形成,主要表现为无定形结构。硫磺素T荧光分析:DSS组对Aβ纤维形成的抑制率为91.38%。结论提示DSS在体外有抑制Aβ聚集和纤维形成的作用。Objective To investigate if Danggui Shaoyao San could inhibit deposition of amyloid-β-protein (Aβ). Methods The solution in aging group was Aβ1-42. peptide. The solution in DSS group was Aβ1-42 peptide and DSS. The solution was incubated for 7 days in 2 group. The structure of Aβ fibril was observed with transmission electron microscopy and thioflavine-T fluorometric assay. Results The solution in aging group showed a high density of fibril Aβ aggregation and formation. However, only a few fibrils were found in DSS group by transmission electron microscopy. To compare with aging group, the Aβ fibril formation and aggregation in the solution of DSS group was inhibited 91.38% by thioflavine-T fluorometric assay. Conclusion The results showed that DSS could inhibit Aβ fibril formation and aggregation to a certain extent in vitro. It suggest that DSS may play a role in the drug therapeutic of Alzheimer's disease by interfereing with the deposition of amyloid-β-protein.
