摘 要:目的探讨关节盘移位后髁突软骨中转化生长因子beta1(transforminggrowthfactorbeta1,TGFβ1)表达与分布的变化规律及可能意义。方法22只日本大耳白兔,16只用于建立关节盘前移位的动物模型,分别于术后1、2、4、8周处死,4只行模拟手术作为手术对照组,另2只不行手术作为空白对照。免疫组化法检测髁突软骨中TGFβ1的表达与分布。结果实验组关节盘成功前移位。对照组TGFβ1主要表达于髁突肥大层及钙化软骨层,实验组关节盘移位后,过渡层及增殖层中TGFβ1表达逐渐增强。8周时髁突软骨全层细胞几乎都表达TGFβ1。结论关节盘移位后TGFβ1的分布及含量发生了改变,这种改变与关节盘前移位病程进展一致。提示TGFβ1可能介导了关节盘移位后疾病发展过程中机械刺激对髁突的改建作用。Objective To evaluate the expression of transforming growth factor-beta 1 (TGF-β1) in cartilage of rabbit mandibular condyle after anterior disc displacement (ADD). Methods Twenty-two healthy adult Japanese rabbits weighing from 2.5 to 3 kg were used in this study. Sixteen rabbits were subjected to surgical anterior disc displacement in the right temporomandibular joints, every 4 rabbits were sacrificed 1, 2, 4, 8 weeks post-operation, 4 rabbits were used as sham-operative control and the other two without operation were used as normal control. The expressions of TGF-β1 in cartilage of mandibular condyle were studied by S-P immunohistochemical staining. Results Experimental and sham-operative animals tolerated the surgical procedures well. The discs displaced anteriorly were observed successfully in the experimental rabbits by general and microscope observation. In control group,. TGF-β1 was mostly expressed in the hypertrophic cartilaginous layer and calcified cartilaginous layer. After anterior disc displacement, the TGF-β1 expression in transitional layer and proliferative layer was increased. In 8 weeks group after ADD, cells in all layers of mandibular condyle almost expressed TGF-β1. Conclusion After ADD, the expression of TGF-β1 in mandibular condyle changed, which was consistent with the process of ADD developed. It suggested that TGF-β1 might regulate the remodeling of the mandibular condyle induced by the mechanical stimulation in the disc displacement process.
