发育不同时期大鼠下丘脑视前区前胸腺素α1-mRNA表达水平变化及基因序列分析

摘  要:目的研究胸腺素在大鼠发育不同时期在下丘脑的表达和下丘脑胸腺素的基因序列。方法运用RT-PCR定量检测和基因序列分析技术,分别检测胚胎12天、14天、16天、19天、出生当天、生后30天、60天SD大鼠下丘脑前胸腺素α1-mRNA的表达。结果从SD大鼠下丘脑视前区扩增出的前胸腺素α1-mRNA的特异性条带,序列分析结果表明,其PCR产物的基因序列与文献报道大鼠胸腺来源的完全一致;大鼠下丘脑视前区前胸腺素α1-mRNA的表达水平随大鼠发育的成熟而呈递减趋势。结论胚胎12天到生后60天大鼠下丘脑视前区表达与胸腺来源完全一致的前胸腺素α1 mRNA,随着大鼠发育成熟而逐渐减少,提示胸腺素α1在个体发育过程中可能参与大鼠下丘脑的功能调节。Objective To explore the expression and gene sequence of thymosin in the hypothalamus of the rats in the different periods of development. Methods The expression and sequence of prothymosin alphal (PT-αl)- mRNA was analyzed by quantitative RT-PCR and sequencing technique in the hypothalamus of SD rats on the embryonic days El2, El4, E!6, El9, and the postnatal days P0, P30, P60. Results The specific PCR products of PT-α1-mRNA were amplified from the preoptic area of the rat hypothalamus, and the amplicons were sequenced identical to the reported sequence of PT-α1-mRNA of rat thymus. Furthermore, the expression level of PT-α1-mRNA in the preoptic area of the rat hypothalamus decreased with the maturation of the rats. Conclusion PT-α1 mRNA, expressed in the preoptic area of the hypothalamus, was identical completely to thymus originated PT-α1 mRNA with the decrease of its level from E12 to P60, suggesting that thymosin-α1 might be involved in the regulation of the functions of rat hypothalamus during the ontogenetic process.
