摘  要:目的建立高效液相色谱法-蒸发光散射检测器(HPLC-ELSD)测定苦杏仁中苦杏仁苷含量的的分析方法。方法采用Hypersil-ODS(4.0mm×250mm,5μm)色谱柱;流动相为甲醇-水(30∶70),流速0.5ml/min;柱温25℃;利用ELSD检测器检测:漂移管温度98℃,载气流速3.2L/min。结果苦杏仁苷进样量在1.0 ̄15.1μg范围内,标准曲线呈良好的线性关系,r=0.9999,苦杏仁苷的平均回收率为99.0%,RSD为2.9%(n=5)。结论本方法简便快速,结果准确可靠,为苦杏仁苷的含量测定提供了一个新方法,可用于苦杏仁药材及含苦杏仁复方制剂的质量控制。High-performance liquid chromatography with evaporative light-scattering detection (HPLC-ELSD) was employed for determination of amygdalin content in Semen Armeniacae Amarum. The detection was performed with the column of Hypersil-ODS (4.6 mm×250 ram, 5μm) and column temperature of 25 ℃. The mobile phase was methanol-water (70:30) with flow rate of 0.5 ml/min. Evaporative light-scattering detector was used and the drift tube temperature was set at 98℃ with the gas flow rate of 3.2 L/min. A standard curve was generated, which was linear in the range of 1.0-15.1 μg for amygdalin content (r=0.999 9). The average recovery of amygdalin was 99.0% with RSD of 2.9% (n=5). Besides simplicity and rapidness, the method yields accurate and reproducible results and can therefore be used in the quality control of Semen Armeniacae Amarum.
