摘  要:目的探讨重组人肿瘤坏死因子α(rhTNFα)对血管内皮细胞凝血、纤溶功能的影响,脑泰方提取物(NTE)对其的作用。方法将融合生长的人脐静脉内皮细胞(HUEC)随机分为空白对照组、NTE对照组、rhTNFα组和NTE高、中、低剂量组,检测100 ng/mL的rhTNFα作用24 h后内皮细胞冻融液的促凝活性、条件培养液中血管性假血友病因子(vWF)含量和组织型纤溶酶原激活物(tPA)、纤溶酶原激活物抑制物(PAI)的活性。结果与空白对照组比较,NTE对照组vWF含量和tPA活性变化无显著性(P>0.05)、PAI活性和促凝活性显著下降(P<0.01),rhTNFα组促凝活性、PAI活性以及vWF含量显著升高(P<0.01),tPA活性变化无显著性(P>0.05);与rhTNFα组比较,NTE各剂量组促凝活性和PAI活性显著降低(P<0.05或P<0.01),NTE中、高剂量组vWF含量均明显减少(P<0.05或0.01),NTE低剂量组vWF含量减少无显著性(P>0.05);NTE对rhTNFα诱导的促凝活性、vWF含量及PAI活性升高的抑制作用均具有明显的剂量效应关系(r分别为-0.83、-0.85、-0.76,P<0.05)。结论NTE能显著抑制正常内皮细胞的促凝活性和PAI活性,对rhTNFα诱导内皮细胞促凝活性、PAI活性及vWF含量的升高均有较好的抑制作用,提示脑泰方对血管内皮细胞抗血栓功能具有较好的调节作用。Objective To observe the functional changes of coagulation and fibrinolysis of human umbilical veins endothelial cells (HUECs) induced by recombined human tumor necrosis factor alpha (rhTNFa) and the effect of Naotaifang Extract (NTE) on it. Methods The cultured HUECs were randomized into six groups: control group, NTE control groups(only 2.0 mg/mL NTE), rhTNFa group ( 100 ng/mL rhTNFa) and 3 doses NTE groups( 100 ng/mL rhTNFa and 6.0 mg/mL or 2.0 mg/mL or 0.67 mg/mL NTE). The coagulation activities of frozen-dissolved HUECs, von willebrand factor (vWF) contents in the conditioned-media and tissue-type plasminogen activator(tPA) and its inhibitor(PAI) activities were detected after 24 hours. Results To compare with the control group, the changes of vWF contents and tPA activities had no significance( P 〉 0.05) and the activities of coagulation and PAl markedly reduced( P 〈 0.01) in the NTE control group, the activities of coagulation and PAI and the contents of vWF were significantly elevated( P 〈 0.01) and the tPA activities didn't have marked change( P 〉0.05) in the rhTNFα group. To compared with the rhTNFa group, the activities of coagulation and PAI in all 3-dose NTE groups the vWF contents in high-and middle-dose groups significantly decreased ( P 〈 0.05 或 P 〈 0.01 ), and the decrease of vWF contents in low-dose NTE group was not significant( P 〉 0.05). The inhibiting effect of NTE on the hoist of the coagulant activities, the vWF contents and the PAI activities had marked dose-effect relation(r was re- spectively -0.83, -0.853 and -0.755, P 〈0.05 ). Conclusions NTE could be effective to inhibit the activities of coagulation and PAI of the HUECs non-stimulated and stimulated by rhTNFα. It suggested that NTE could be effective to adjust the anti-thrombesis function of vasenlar endothelial cell.
