涤痰汤对大鼠脑出血后血肿周围组织中活化caspase-3表达的影响

摘　　要：目的探讨大鼠脑出血后血肿周围神经元活化caspase—3的表达及涤痰汤的干预作用。方法用Ⅶ胶原酶脑内立体定位注射诱导大鼠脑出血模型,免疫组织化学法检测神经元线粒体内caspase—3的释放情况。结果Ⅶ胶原酶造模后模型组血肿周围于6 h出现阳性表达细胞,1 d阳性细胞计数达高峰,3 d出现轻度下降,5 d明显下降。与假手术组比较差异明显(P<0.01)。涤痰汤组与同时间模型组比较在6 h1、d计数无明显差异,35、d时阳性细胞减少明显(P<0.01)。结论大鼠脑出血后血肿周围神经元活化caspase—3表达明显上调;涤痰汤能减少活化caspase—3的表达,阻止神经元的凋亡。Objective To observe the expression of activating caspase-3 around hematoma after intracerebral hemorrhage（ICH） in rats and the effects on interference of Ditan decoction （DID）. Methods Immunohistochemical methods were used for examming the expression of activated caspase-3 around hematoma after ICH in rats. Results The activated caspase-3 positive cells appeared around the hematoma at 6 hours after operation, and then, got to the peak at 1 day, descend slightly at 3 days and descend distinctly at 5 days after operatiou. Compared with the sham operation group, it varied evidently（ P 〈 0.01）. The number of positive cells of the DID group was obviously smaller than that of the model group at 3 d,5 d（ P 〈 0.01） ,but not at 6 h and 1 d. Conclusion The activated caspase-3 increase obviously in the neuron around the hematoma after ICH; DID can markedly depress the expression of activated caspase-3 and prohibit neuronal apoptosis.
