活血通窍方对大鼠实验性变应性鼻炎的一般行为及组胺的影响

摘　　要：目的研究活血通窍方经鼻腔给药对大鼠变应性鼻炎的一般行为及组胺的影响。方法采用2%卵蛋白(OVA)生理盐水溶液喷雾吸入法,造成大鼠变应性鼻炎模型。取造模成功动物50只,随机分为5组,即中药大剂量组(4 g.kg-1.d-1)、中药中剂量组(3 g.kg-1.d-1)、中药小剂量组(2 g.kg-1.d-1)、西药组及模型组。于造模第11天起,各组均每日滴鼻给药3次,每侧15μl,模型组及正常对照组用等容量的生理盐水滴鼻治疗。西药用伯克纳鼻喷雾剂,每次每侧1揿。连续用药15 d。观察给药后大鼠鼻部外在行为学的改变情况;用荧光比色法测定鼻黏膜组织中的组胺含量;观察鼻黏膜切片的病理学改变。结果活血通窍方各剂量组均有改变大鼠行为的作用,能显著减轻变应性鼻炎所致的鼻黏膜组织形态学的改变,与模型组相比有显著意义(P<0.05);活血通窍方大剂量组和中剂量组能抑制鼻黏膜中的组胺含量,与模型组相比有显著意义(P<0.05),结论经活血通窍方滴鼻给药后,显示了药物对变应性鼻炎模型的作用。其作用机制可能与抑制嗜酸细胞活性、抑制炎症介质的释放有关。Objective To investigate the therapeutic effects of Huoxue Tongqiao Decoction （HTQD）on rats with allergic rhinitis. Methods Fifty rats were randomized into five groups （normal group, group treated by Beelomethasone Dipropionate Nasal Spray, and groups treated by High, middle and low dose of HTQD）. Behaviour, histamine content in nasal mucosa, morphological features of nasal mucosa under light microscopy were observed. Result Compared with model group, HTQD relieved the pathological symptoms of allergic rhinitis and morphological changes of nasal mucosa, meanwhile, high and middle dose of HTQD decreased the release of histamine in nasal mucosa. Conclusion HTQD has significant effect-against allergic rhinitis. The mechanism relates possibly to its effects of resisting the releasing of inflammatory factor and inhibiting the activity of eosinophil.
