摘  要:目的观察在正常和活动力低下的人精液精子中白细胞介素-6(IL-6)及其受体(IL-6R)和GP130的表达差异。方法收集活动力正常的人精液精子和A、B级精子低于20%的弱精子症患者的精液精子,采用逆转录-聚合酶链反应 (RT-PCR)和免疫细胞化学的方法检测IL-6及其受体和GP130的表达水平。结果与活动力正常的精液精子比较, GP130在活动力低下患者精液精子的表达显著降低(P<0.01),本实验未检测到IL-6和IL-6R在人精液精子的表达。结论精液精子GP130的表达减少与人精子活动力低下相关,提示GP130表达减少可能是精子活动力低下的原因之一。Objective;Interleukin-6 (IL-6) shows extensive functions in the process of physiology and pathophysiology by binding to IL-6 receptor (IL-6R) and GP130. The present study is to compare the expression of IL-6, IL-6R and GP130 in ejaculated spermatozoa from healthy volunteers and the patients with asthenospermia. Methods. Semen samples were obtained from the healthy volunteers and the patients with asthenospermia from the Center for Reproductive Medicine, Peking University Shenzhen Hospital. Semen analysis was performed according to WHO criteria. The spermatozoa were collected and purified by the Percoll gradient technique to separate seminal plasma and to remove cell contamination. RT-PCR and immunocytochemistry were used to detect the expression of IL-6, IL-6R and GP130 in spermatozoa from two groups. Results. The mRNA expression of GP130, but not IL-6 and IL-6R, was detected by RT-PCR in human ejaculated spermatozoa. Compared with the healthy volunteers, the patients with asthenospermia showed a significant decrease in the expression of GP130 mRNA. To further confirm the results from RT-PCR, immunocytochemistry was used to investigate the protein expression of GP130 in human ejaculated spermatozoa. The results showed that the expression of GP130 in the patients with asthenospermia was significantly lower than that in healthy volunteers, which was similar to the results from RT-PCR. Conclusion. The expression of GP130 in ejaculated spermatozoa is significantly decreased in the patients with asthenospermia, which suggests one possible reason for the low sperm motility of the patients.
