摘 要:目的探讨重组人白血病抑制因(LIF)对人早孕滋养层细胞着床相关分子原癌基因c-jun、c-fos表达的影响及其作用环节。方法将人早孕滋养层细胞培养于DMEM培养液中,加入不同浓度LIF(1、10和100 ng)作用1 h后,分别提取总RNA;逆转录-聚合酶链反应(RT-PCR)方法检测原癌基因c-jun和c-fos的表达。结果正常培养的人早孕滋养层细胞有一定水平的c-jun、c-fos基因表达;LIF对人早孕滋养层细胞c-jun、c-fos的表达均有上调作用,c-jun、c-fos mRNA水平增加1.5~3倍,并且调节呈剂量依赖性。结论LIF可能通过调节滋养层细胞原癌基因c-jun和c-fos表达而影响胚胎的植入。Objective: Leukemia inhibitory factor (LIF) is essential for embryo implantation. The purpose of this study is to understand the effects of LIF on the expression of c-jun and c los in human trophoblast cells. Methods: Trophoblast cells were isolated from placenta villi of health women of legal abortions during the first trimester of pregnancy and cultured in DMEM medium. After the cells were treated with different concentrations of LIF (1 ng, 10 ng and 100 ng, respectively) for 1 h, the expression of c-jun mRNA and c-fos mRNA were detected with reverse transcription-polymerase chain reaction (RT-PCR). Results: Trophoblast cells expressed both of c jun and c-fos mRNA in vitro. Treatment of LIF signifi- cantly increased the levels of c-jun and c-los in trophoblast cells (P〈0.01). The increases of c-jun and c-los were 1.5 to 3 folds. In addition, the stimulation of c-jun 'by LIF was in a dose dependent manner. Conclusion: These data suggest that LIE is probably involved in embryo implantation by stimulating the expression of c jun and c-los.
