摘  要:目的:探讨益气活血颗粒剂对纤维化肺组织TGF-β1、TNF-α表达的影响。方法:20只Wistar雄性大鼠随机分为模型组、中药治疗组、激素组及正常对照组4组,采用BLEMA5建立大鼠肺间质纤维化模型,中药治疗组给予益气活血颗粒剂(250mg/kg)灌胃,激素组应用氢化考的松(25mg/kg)肌注。观察肺组织HE、VG染色病理改变,采用核酸原位杂交法检测TGF-β1、TNF-α表达量。结果:模型组肺泡炎和肺纤维化程度较重,且其胶原纤维沉积较多,TGF-β1、TNF-α的表达增多;而中药治疗组肺泡炎和纤维化程度较轻,胶原沉积明显减少,TGF-β1、TNF-α表达减少。激素组纤维化程度比模型组减轻,但比中药组重,TGF-β1、TNF-α表达减少不明显。结论:益气活血颗粒剂通过下调TGF-β1、TNF-α的表达而明显减轻了肺间质纤维化程度。Objective:To investigate the effect of Yiqihuoxue (YQHX) on TGF-β1 and TNF-αexpressions in lung tissues in bleomycin-induced pulmonary fibrosis model in rats. Methods: Animal model of bleomycinA5-induced pulmonary fibrosis was established by BLEMA5 in rats. 20 Wistar rats were divided into normal control, fibrosis, treatment and corticosteroids groups. Normal control group was given 0.9% saline (2 ml/kg) intratracheally by instillation, fibrosis group bleomycinA5(5 mg/kg)intratracheally by instillation, treatment group YQHX 1,2,3 (250 mg/kg) daily by oral , and corticosteroids group hydrocortisone (25 mg/kg im) daily. All rats were killed on 28 days. The HE and the VG stain were pathologically examined and the expression of TGF-β1 and TNF-α mRNA in the lung tissues were analyzed by nucleic acid hybridization in situ. Results:YQHX1,2,3 obviously alleviated rats alveolitis and fibrosis. TGF-β1 and TNF-α levels were higher in fibrosis group than those in normal control and treatment group (P〈0.01). Conelusion:YQHX1,2,3 can treat pulmonary fibrosis through reducing the expression of TGF-β1 and TNF-α in lung tissues.
