摘 要:目的:合成球状单分散麻黄碱分子印迹聚合物并研究印迹聚合物的识别特性。方法:采用多步溶胀和悬浮聚合相结合的方法,以麻黄碱为模板分子,甲基丙烯酸和二甲基丙烯酸乙二醇酯分别为功能单体和交联剂,合成麻黄碱印迹聚合物微球。通过 HPLC 研究微球的识别特性和色谱条件对分离的影响。结果:麻黄碱与伪麻黄碱以及结构类似物在印迹柱上得以分离,非印迹柱则没有这种分离能力。流动相中醋酸比例增加使得麻黄碱保留减小,进样量对峰形有较大影响。结论:印迹聚合物对模板分子具有很强的亲和力和特定的选择性。合成的麻黄碱分子印迹柱有较大的结合容量,可应用于生物样品中麻黄碱RP-HPLC 检测的预处理柱和麻黄草中麻黄碱的提取。Objective:To prepare ephedrine-imprinted polymeric microspheres in an aqueous system and to study the recognition mechanism. Method:Using methacrylic acid(MAA) as functional monomer,and ethylene glycol dimethacrylate(EDMA) as cross-linker, ephedrine-imprinted polymeric microspheres was prepared by multi-step swelling and suspension polymerization. The binding properties were studied by HPLC. Results:Stable macroporous polymer beads with good size monodispersity were obtained,and the average size of which was 4. 5 m. It was found that the baseline separation of the analogues was achieved on a short column. The separating degree was good ,when the chromatographic condition were as follows : acetonitrile-acetic acid (99. 8: 0. 2, v/v) as the mobile phase, sample amount 40 g, flow rate 1.0mL min^- 1, and column temperature at room temperature respectively. Conclusion :The prepared polymers can be used as separation medium of ephedrine with good specific recognition ability and high binding capacity.
