摘  要:目的观察石杉碱甲对血管性痴呆小鼠海马神经元胆碱乙酰转移酶及其mRNA变化的影响,以探讨两者在血管性痴呆发病中的作用及石杉碱甲对其影响的机制。方法双侧颈总动脉线结,反复缺血-再灌注法制备模型,药物组用石杉碱甲溶液灌胃,跳台试验和水迷宫试验观察其行为学改变,采用免疫组织化学和原位杂交技术观察小鼠海马神经元胆碱乙酰转移酶及其mRNA的表达变化。结果石杉碱甲明显改善了模型小鼠学习、记忆成绩(P<0.01)。同时,药物组海马的胆碱乙酰转移酶阳性神经元的面密度值为(0.1133±0.0118),高于模型组(0.0827±0.0118);并且药物组和模型组海马胆碱乙酰转移酶mRNA阳性神经元的面密度值相比,前者(0.1122±0.0137)也高于后者(0.0798±0.0184)(P<0.01)。而海马两者的表达在药物组与假手术组间差异无显著性。结论石杉碱甲改善血管性痴呆小鼠学习、记忆成绩与其恢复海马低水平的胆碱乙酰转移酶及其mRNA有关。Objective To observe the effect of .huperzine A on choline acetyltransferase in neuron of hippocampus in mice with vascular dementia. Methods The mice were subjected for ischemic reperfusion repeatedly on bilateral common carotid arteries by knots to establish the VD models. The behavioral abnormalities were investigated by step-down test and water maze test. Huperzine A was given to the mice in medication group, choline acetyltransferase in neuron of hippocampus were observed by immunohistochemistry technique. Results The grades of learning and memory in VD mice were improved by huperzine A. Surface density value of positive choline acetyltransferase neuron of hippocampus in drug group(0.1133 ± 0.0118 )was more than in model group( 0.0827 ± 0.0118 ). And surface density value of positive choline acetyltransferase mRNA neuron of hippocampus in drug group ( 0.1122 ± 0.0137 ) was also more than that in model group ( 0. 0798 ± 0.0184 ) ( P 〈 0.01 ). However, no difference was discovered between drug group and sham group. Conclusion Huperzine A improved dysfunction of learning and memory in VD, which suggested that huperzine A restored expression of choline acetyltransferase.
