摘 要:目的研究杜仲原药材中绿原酸的含量测定方法。方法采用高效液相色谱(HPLC)法,对杜仲中绿原酸提取时的提取溶剂、方法、时间和次数进行了优选。以LichrospherC18(4.6mm×250mm,5μm)为分析柱,流动相为乙腈-水-磷酸溶液(13∶86.5∶0.5);柱温:25℃;检测波长:327nm;流速:0.8mL/min,采用面积外标法。结果杜仲粉用体积分数为60%甲醇水溶液回流提取2次(每次100mL,第1次1.5h,第2次0.5h),可提出杜仲原药材中绿原酸总量的95.68%。线性范围2.22×10-2~55.40×10-2μg,平均回收率为99.55%,RSD=1.98%(n=6)。结论本法简便、灵敏、重现性好,可用于测定杜仲中绿原酸的含量。Objective The methods for determination of Chlorogenic Acid(CA) in Eucommia ulmodies Oliv were established and determinated the content of CA. Extract solvent, method, time and times were studied and HPLC method for determination was used. Methods The HPLC system consisted of the column of Lichrospher C18 (4.6 mm×250 mm, 5μm) and acetonitrtle-water-phosphoric acid (13 : 86.5 : 0.5) as the mobile phase. The temperature was 25℃. The detective wavelength was 327nm. The flow rate was 0.8 mL/min. External standard method was used. Results The powder of eucommia was extracted twice with 60%methanol-water solution at 100 ~C, the first time for 1.5 h,the second for 0.5 h was the optimum one. The method was linear within the range of 2.22×10^-2-55.40×10^-2 μg, the average recovery was 99.55%, and the RSD was 1.98% (n=6). Conclusion The method is simple, sensitive and highly reproductive, and it is suitable for ananlysis of CA in Eucommia ulmodies Oliv.
