五灵胶囊对四氯化碳D-氨基半乳糖与内毒素所致肝损伤影响

摘　　要：目的探讨五灵胶囊对慢性肝损伤的保护作用与机理。方法采用CCl4与DGlaN诱导小鼠慢性肝损伤模型以及体外DGlaN、LPS诱导大鼠原代肝细胞损伤模型,酶法测定血清、肝组织、细胞培养上清ALT、MAO、LDHL、GSHST、MDA、CHE等指标和病理形态学检查。结果五灵胶囊能显著降低由CCl4、DGlaN诱导小鼠肝损伤时所致高血清与组织ALT水平,明显升高血清与组织CHE;可使DGlaN、LPS损伤原代肝细胞培养上清MAO、LDHL、GSHST、MDA水平显著降低,受损的肝细胞修复;病理形态学结果也表明其对慢性肝损伤的治疗作用。结论五灵胶囊能有效地对抗由CCl4的脂质过氧化作用所致的肝细胞坏死,能显著阻滞DGlaN诱导小鼠慢性肝损伤,明显地降低DGlaN、LPS诱导损伤原代大鼠肝细胞。五灵胶囊治疗慢性肝损伤作用与保护肝质膜,增加肝细胞膜的稳定性,促进肝细胞内蛋白合成,有效地防止膜的脂质氧化作用有关。Aim to investigate the effect and mechanism of Wuling capsule on chronic liver injuries. Method The liver damage model of mice induced by CC14 and D-GlaN actosamine （D-GlaN） was used.The model of the rat prototype liver cell was adopted induced by D-GlaN and LPS in vitro. The ALT, MAO, LDH-L, GSH-ST, MDA, CHE in serum, hepatic tissue,cell culture fluid, and the histopathology were observed at the same time. Result WuLing capsule could inhibit hep- atic high ALT in the serum and tissue induced by CC14, D-GlaN. The CHE was increased in the serum and tissue. The MAO, LDH-L, GSH-ST and MDA in rat liver prototype cells damaged by CC14 and D-GlaN were decreased significantly. Injuried liver cells were restored. Effect of WuLing capsule on the chronic liver injuries were showed by the histopathology. Conclusion Wuling capsule could effectively restore necrotic liver cells induced by lipid-peroxide of CC14, block chronic liver trauma in mice caused by D-GlaN, and restore the lesion of prototype hepatic cells induced by D-GlaN or LPS. The effect of Wuling capsule on the chronic liver lesion correlated with the ability to protect hepatic membrane, increase the stability of hepatic-cellular membrane, promote the synthesis of the protein in liver cell, and prevent effectively the effect of membrane lipid-peroxide.
