摘  要:目的研究外源性胰岛素原C肽对2型糖尿病患者离体红细胞膜Na+-K+-ATP酶活性及红细胞内Ca2+浓度的影响。方法取2型糖尿病患者空腹血,肝素抗凝,分离红细胞,生理盐水重新悬浮,加外源性C肽,分别测定细胞膜上的Na+-K+-ATP酶活性和细胞内Ca2+浓度。结果外源性C肽的加入可升高2型糖尿病患者红细胞膜的Na+-K+-ATP酶活性,并降低红细胞内的Ca2+浓度。结论胰岛素原C肽可以改善2型糖尿病患者降低的红细胞膜Na+-K+-ATP酶活性。Objective To study the effect of human proinsulin C-peptide on erythrocyte Na^+-K^+-ATPase activity and Ca^2+ concentration in patients with type Ⅱ diabetes mellitus in vitro. Methods The empty stomach blood was taken out from the patients with type II diabetes mellitus in the morning and heparin was used as anticoagulant agent at the same time, then the erythrocyte was separated and resuspended in the equal volume normal saline. After the exogenous proinsulin C-peptide was added into the suspended erythrocyte in saline, the erythrocyte Na^+-K^+-ATPase activity and the Ca^2+ concentration were measured. Results The activity of erythrocyte membrane Na^+-K^+-ATPase was improved and the concentration of Ca^2+ in erythrocyte was decreased after the exogenous proinsulin C-peptide was added in vitro. Conclusion The decreased activity of erythrocyte membrane Na^+-K^+-ATPase in patients with type Ⅱ diabetes mellitus could be improved after treated with proinsulin C-peptide in vitro.
