摘 要:目的探讨湖北省流感病毒分离株的变异、进化发展过程,比较这些毒株之间的遗传进化亲缘关系,探讨湖北省流感病毒血凝素基因变异规律,为湖北省流感防治提供理论基础.方法对省内1983~2002年分离到的8株H3N2亚型流感病毒HA1区基因进行序列测定,并用生物信息学研究方法,分析比较各年代病毒分离株的进化关系.结果随着时间的变化,湖北省内流感病毒HA1基因间核甘酸替换数增加,随之抗原决定簇A和B的氨基酸有改变.结论分析1983~2002年8株流感病毒株,根据其HA1基因序列,按年代特征划分为两个谱系.其中一个谱系是1985年及其以前分离的毒株.上个世纪90年代后分离的毒株为一个谱系,90年代及2002年的毒株在进化树上和80年代处于不同分支.Objective To study the characteristics of HA1 gene of influenza viruse subtype A3 isolated in Hubei Province during 1983-2002 and evaluate its variation and evolution. Methods 8 strains of influenza viruses were inoculated in eggs and the RNA of these viruses were extracted with RNeasy Mini kit. The genes of HA1 were amplified by RT-PCR . The PCR products were sequenced by sequence equipment (ABI- 377) . All data were analyzed by the software DNA star 4. 0. Results The substituting number of nucleotides of HA1 gene were increasing with time and the amino acids changed in A and B regions of the antigenic determinants. Conclusion It showed that the H3N2 influenza virus strains were divided into two major lineages. One lineage included the strains before 1985 and the other lineage included the strains isolated in 90's and 2002. The two lineages were different in phylogenetic tree.
