摘 要:目的研究大鼠血清对猪冠状动脉平滑肌细胞(SMC)钙激活钾通道(KCa)的作用,为血清药理学方法与膜片钳技术结合应用于相关离子通道机制研究奠定基础。方法用EGTA处理血清中的Ca2+、Mg2+等离子,用膜片钳单通道技术研究处理后的血清对SMC上的KCa的影响。结果处理后的血清浓度依赖性的不可逆性的阻断SMC上的KCa。在细胞贴附式膜片(cell-attached patch)下,血清10、50、100、150μl·ml-1,使通道开放概率从0.0285±0.0102分别降低到0.0205±0.0046(n=7,P>0.05)、0.0145±0.0007(n=7,P<0.05)、0.0045±0.0002(n=7,P<0.01)、0(n=7,P<0.01)。在内面向外式膜片(inside-out patch)下,血清10、50、100、150μl·ml-1,使通道开放概率从0.3821±0.0491分别降低到0.1298±0.0331(n=7,P<0.05)、0.0405±0.0048(n=7,P<0.05)、0.0117±0.0011(n=7,P<0.01)、0.0008±0.0005(n=7,P<0.01)。随着血清浓度的增加,通道活动逐步减少至完全抑制,这种作用冲洗后不可逆。结论血清对猪冠状动脉平滑肌细胞钙激活钾通道有一定阻断作用。Objective: To study the action of serum of SD rat on calcium - activated potassium channels (KCa) in porcine coronary artery smooth muscle cells (SMC) and to establish the foundation for combining serum pharmacology with patch clamp technique in studying ion channels. Methods: Ca^2+ , Mg^2+ in serum were treated with EGTA. The effect of the treated serum on KCa in SMC was observed using patch clamp single channel technique. Results: Serum could block KCa in porcine coronary artery smooth muscle cells in a dose dependent manner, and it was not reversible. The open probability (P0) of the channels was reduced from 0.0285 ± 0.0102 to 0.0205 ± 0.0046(n = 7, P 〉 0.05) ,0.0145 ± 0.0006(n = 7, P 〈 0.05) ,0.0045 ± 0.0002(n = 7, P 〈 0.01) and 0(n = 7, P 〈 0.01)on the cell - attached patch when adding serum of 10,50,100,and 150μl·ml^-1 ; The open probability (P0) of the channels was reduced from 0.3821±0.0491 to 0.1298±0.0331(n=7, P〈0.05),0.0405±0.0048(n=7, P〈0.05),0.0117±0.0011(n=7, P〈 0.01) ,and 0.0008±0.0005(n = 7, P 〈 0.01)on the inside - out patch when adding serum of 10,50,100 and 150μl·ml^-1. With the dose of serum increased, the activation of the channels was inhibited until it was completely blocked. The effect was not reversible after washout. Conclusion: Serum of rat could block KCa in SMC.
