摘  要:目的观察脂质体载体pcDNA3-TGF-β1基因治疗对同种心脏移植物存活的影响。方法大量扩增pcDNA3-TGF-β1,制备脂质体-靶基因混合物及杂交探针,建立同种心脏移植模型,行脂质体载体pcD-NA3-TGF-β1基因转染,观察移植后5d心肌细胞中TGF-β1mRNA表达状况和移植心脏病理改变,观察受体存活情况。结果移植后5d脂质体载体pcDNA3-TGF-β1基因治疗组心肌细胞中TGF-β1mRNA原位杂交阳性表达率为11%,移植心脏排斥反应病理改变明显延缓,同种心脏移植物存活明显延长,但未诱导出长期存活。结论脂质体载体pcDNA3-TGF-β1基因治疗延长了同种心脏移植物的存活。[Objectives] To investigate the impact of liposome-mediated pcDNA3-TGF-β1 gene therapy upon the survival of cardiac allografts. [Methods] After amply the plasmid peDNA3-TGF-β1, the mixture of liposome and target plasmid was prepared and used into the cardiac transplants while the hybridization probes were developed. 5 days post transplanttaion the expression of TGF-β1 mRNA in cardiac muscles was observed by in situ hybridization and the rejection pathological progress were evaluated. The survival of grafts receiving gene therapy was analyzed. [Results] The efficiency of liposome-mediated peDNA3-TGF-β1 gene transfer in cardiac muscle indicated by in situ hybridization was 11% after transplantation in 5th day. The rejection pathological progress was delayed. This therapy prolonged the survival of cardiac grafts significantly but without long-term case. [ Conclusion] The liposome-mediated pcDNA3-TGF-β1 gene transfer can prolong the survival of cardiac grafts significantly.
