益活清胰汤对重症急性胰腺炎大鼠海马CAI区bFGF神经元影响的实验研究

摘　　要：目的探讨益活清胰汤对大鼠重症急性胰腺炎(SAP)合并胰性脑病(PE)海马CAI区bFGF的影响。方法45只SD大鼠建立SAP模型,随机分为治疗组、对照组、正常组,测定6 h1、2 h、24 h各时点血TNF-a、IL-6、ALP2值。对大脑皮层和海马组织形态、嗜碱性成纤维细胞生长因子(bFGF)的表达进行观察。结果B、C两组术后6 h TNF-a、IL-6、ALP2开始升高,24 h达高峰。治疗组6 h1、2 h、24 h各时点的TNF-a、IL-6、ALP2值小于对照组(P<0.01)。海马bFGF表达显著增多。结论益活清胰汤能降低SAP血清TNF-a、IL-6及PLA2水平,改善脑血流量,对抗缺血缺氧,增强SAP时海马组织的bFGF的合成、分泌,阻止神经细胞凋亡和/或促进新的神经元芽及突触形成。Objective： To study the effect of Promote Blood Circulation Qing Yi Decoction on basic fibroblast growth factor （bFGF） neuron in hippocampus CAI position of Rats with severe acute pancreatitis （SAP） united pancreatic encepialopathy （PE）. Methods： Forty - five SD Rats were induced SAP and divided into treatment group, model group and control group. Blood was obtained respectively 6 hours, 12 hours, 24 hours after operation. Expression of TNF - a, IL- 6、 ALP2 was measured. Observe pathomorphologic changes of cerebral cortex and hippocampus, determine the positive basic fibroblast growth factor （bFGF） neuron in hippocampus by immunohistochemical method, study the expression levels of （bFGF） of hippocampus by situ hybridization. Results： In the B group and C group TNF- a,IL- 6,ALP2 began to rise 6 hours after operation, reached the peak 24 hours after the operation. In the treatment group 6 hours, 12 hours, 24 hours after operation, TNF - a, IL- 6 and ALP2 were lower than the model group （ P 〈 0.01 ）. Activities of bFGF in hippocampus ascented by large margin （P 〈 0.01）. Conclusion： promote Blood Circulation Oing Yi Decotion can lower the TNF - a, IL - 6 and ALP2 level of SAP. Regulate brain blood infusion and anti - ischemia, upgrade the level of bFGF in hippocampus inhibited delayed neuron death, stimulating neurons reproduction.
