摘  要:目的:观察缺血再灌注对大鼠心肌细胞凋亡的影响及葛根素对其保护作用,并探讨其可能机制。方法:采用原位末端标记(TUNEL)技术、流式细胞仪方法观察心肌缺血再灌注损伤前后以及使用葛根素后细胞凋亡的情况;用免疫组化方法检测Bc l-2、Bax蛋白表达情况,并在光镜及电镜下观察细胞形态学变化。结果:(1)心肌缺血再灌注损伤后TUNEL阳性细胞数、流式细胞仪中凋亡指数较假手术组增高(P<0.05),使用葛根素后下降(P<0.05);(2)缺血再灌注组Bax、Bc l-2蛋白表达较假手术组增高,加用葛根素后Bc l-2表达增高,而Bax表达则降低。结论:葛根素能抑制心肌缺血再灌注损伤中的细胞凋亡,增加Bc l-2、降低Bax的表达。Objective:To observe the effect of myocardial ischemia- reperfusion on the rat myocardial cell apoptosis and the protective effect of puerarin and to explore the probable mechanism. Methods :The myocardial cell apoptosis was studied respectively by TdT- mediated dUTP nick end -labling( TUNEL), and by flow cyto meter (FCM) in the myocardial ischemia- reperfusion in the groups which used puerarin and in the groups which did not use puerarin, and the expressions of Bax,and Bcl - 2 were determined by using immunohistochemistry, and the changes in morphology were observed by microscope, transmission electron microscope. Results:( 1 )Compared with the sham operated group, the positive cells of TUNEL and apoptotic index of FCM were obviously enhanced in the group of ischemia - reperfusion ( P 〈 O. 05 ) and puerarin could decrease the positive cells and apoptotic index ( P 〈 0. 05 ) ; (2) Compared with the sham operated group, the expression of Bcl - 2 was increased and the expression of Bax was the highest in the group of ischemia - reperfusion ; but the expression of Bcl - 2 became higher and the expression of Bax was decreased after the use of puerarin. Conclusion:Puerarin can inhibit myocardial apoptosis, increase Bcl -2 expression, and decrease Bax expression. Puerarin may play a positive effect in preventing myocardial ischemia - reperfusion injury.
