高效液相色谱法测定石榴皮中鞣花酸的含量

摘　　要：目的建立石榴皮提取物中鞣花酸含量的HPLC测定方法.方法色谱条件Zobax SB C18柱(250 mm×4.6 mm,5μm),柱温30℃,用CH3OH-EtOAc-KH2PO4/H3PO4(0.05 mol/L)(体积比34:2:64)作为流动相,检测波长为254 nm.结果鞣花酸在0.3184～7.96μg范围内与峰面积呈良好的线性关系,r=0.9999.平均加样回收率为98.12%,RSD为1.13%.结论本方法精确可靠,可作为石榴皮药材质量控制的方法.Objective To develop a rapid and sensitive high-performance liquid chromatographic assay for determination of ellagic acid in the extracts of Punica granatum L. pericarp. Method Chromatography was performed on a Zobax SB - C18 column （250 mm×4.6 mm, 5μm） at 254 nm. The mobile phase consisted of a mixture of methanol, ethyl acetate, 0.05 mol/L KH2 PO4 - H3 PO4 （ 34： 2： 64） at a flow rate of 1.0 ml/min. Result and Conclusion The linear range of ellagic acid was 0.3184 - 7.96μg with a correlation coefficient of 0.9999. The average recovery was 98.12% and RSD 1.13%. The method is accurate and reproducible for quality control of Punica granatum L. pericarp.
