摘 要:目的研究麻黄附子细辛汤治疗哮喘的作用机理。方法以IFN-γ代表Th1型细胞因子,IL-4和IL-5代表Th2型细胞因子,以地塞米松为对照,应用ELISA的方法,观察麻黄附子细辛汤及对哮喘患者外周血单个核细胞(PBMC)分泌Th1和Th2型细胞因子的影响和对淋巴细胞凋亡的影响。结果麻黄附子细辛汤孵育后,细胞培养上清液中IL-4、IL-5浓度明显下降,IFN-γ浓度无明显变化,对IL-4的抑制程度随接触时间的延长而增强,并可见到大量凋亡的淋巴细胞。结论麻黄附子细辛汤可能主要是促进Th2细胞的凋亡,并抑制Th2型细胞因子的分泌进而恢复Th1/Th2平衡来发挥抑制哮喘发病的作用的,与激素在临床治疗哮喘方面具有相似的作用机理。Objective To investigate the anti-inflammatory mechanism of Decoction of Ephedra, Aconite and Asarum. Methods PBMC from asthmatic patients were cultured in vitro with the decoction, After 24 and 48 hours cultivation, concentrations of IL-4, IL-5 and IFN-γ in the supernatant were measured with ELISA. And lymphocytes were examined with electron microscopy. ResuhsConcentrations of IL-4 and IL-5 were apparently decreased but IFN-γ showed no obvious change. Moreover, inhibition effect on IL-4 was increased gradually with the exposure time. Observed with electron microscope, mass lymphocyte apoptosis were found. Conclusion The results suggest that this decoction can keep Thl/Th2 balance not only by promoting Th2 cell apoptosis but also by inhibiting Th2-like cytokine secretion. Decoction and dexamethasone have partially similar antl-inflammatory effects on asthma.
