摘 要:目的:通过建立力竭游泳训练模型,观察巴戟天对运动训练大鼠骨骼肌自由基代谢及运动能力的影响。方法:实验于2004-04/12在郑州大学医学院药理教研室完成。选择雄性SD大鼠24只,随机分为安静对照组,过度训练组,过度训练+巴戟天组,每组8只。过度训练组、过度训练+巴戟天组大鼠均采用递增负荷游泳训练建立力竭游泳训练模型,每周训练6d,共7周。过度训练+巴戟天组大鼠同时于训练前1h灌服巴戟天4.5mg/(kg·d)。安静对照组只进行常规游泳训练,30min/d,每周训练6d,共训练7周。7周后将各组大鼠处死,立即分离骨骼肌,用预冷的生理盐水洗净血污,滤纸吸干,按1∶9(W/V)加入低温磷酸盐缓冲液(pH7.2)匀浆,离心(5000r/min,25min,0~4℃),取上清液,在1~4℃的冰箱保存,以测定超氧化物歧化酶和谷胱甘肽过氧化物酶活性、Ca2+-ATP酶和Na+-K+-ATP酶活性。组间均数比较采用t检验。结果:各组大鼠均进入结果分析,无脱落。过度训练组大鼠力竭时间明显低于安静对照组和过度训练+巴戟天组(P<005);经过7周训练,过度训练组大鼠肌细胞超氧化物歧化酶和谷胱甘肽过氧化物酶活性、Ca2+-ATP酶和Na+-K+-ATP酶活性显著低于安静对照组和过度训练+巴戟天组(P<005),安静对照组与过度训练+巴戟天组无显著性差异。结论:巴戟天可显著提高运动大鼠抗自由基氧化的功能,使大鼠运动能力明显增强。AIM: To observe the effects of Radix Morindae Officinalis (RMO) on the free radical metabolism of skeletal muscles and exercise performance in rats of exercise training by establishing models of exhausted swimming training. METHODS: The experiment was carried out in the Department of Pharmacology, Medical College of Zhengzhou University from April to December 2004. Twenty-four male SD rats were randomly divided into 3 groups with 8 rats in each group: quiet control group, overtraining group, overtraining+RMO group. Rats in the overtraining group and overtraining+ RMO group were made into models of exhausted swimming training by progressive increase of loaded swimming training, 6 days a week for 7 weeks. In the overtraining+RMO group, the rats were perfused by RMO (4.5 mg/kg per day) at 1 hour before training. In the quiet control group, the rats were only force to swim routinely, 30 minutes a day, 6 days a week for 7 weeks. All the rats were killed after 7 weeks to separate the skeletal muscle immediately, the blood stain was cleaned with the precooled physiological saline, died with the filter paper. According to 1:9 (W/V), the low temperature phosphate buffer solution was added (pH7.2) to be homogenization, centrifugated (5000 r per minutes, 25 minutes, 0 - 4 ℃), the supernatant was fetched, kept in 1 - 4 ℃ refrigerators, the activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), Ca^2+- ATPase and Na^+-K^+-ATPase were determined. RESULTS: All the rats entered the analysis of results without deletion. The time of exhaustion in the overtraining group was obviously lower than those in the quiet control group and overtraining group+RMO (P 〈 0.05). After 7-week training, the activities of SOD, GSH-Px, Ca^2+-ATPase and Na^+- K^+-ATPase in the overtraining group was obviously lower than those in the quiet control group and overtraining group+RMO (P 〈 0.05), and there were no insignificant differences between the quiet control
