摘  要:目的研究不同频率张应变对血管平滑肌细胞表型转换的影响。方法应用FX-4000T细胞应变加载系统,对体外培养的大鼠血管平滑肌细胞施加10%应变,频率分别为0.5 Hz1、Hz和2 Hz,加载时间12 h和24 h后,采用Real time RT-PCR、Western blot、免疫荧光化学等技术检测不同频率张应变下血管平滑肌细胞的αlpha-肌动蛋白(-αactin)的表达变化。以未加载张应变的血管平滑肌细胞为对照组。结果血管平滑肌细胞的-αactin的蛋白和RNA表达量在1 Hz条件下比相同时相点的其他频率组均高。结论不同频率的张应变可以影响血管平滑肌细胞-αactin的表达量,提示应变频率的改变可能参与血管平滑肌细胞表型转换的调节。Objective To investigate the effect of different frequencies of cyclic tensile strain on phenotype transformation of vascular smooth muscle cells ( VSMC ). Methods Aortic vascular smooth muscle cells of rats grown on dishes coated with collagen I were subjected to 10% elongation and various frequencies of mechanical strain using the Flexereell 4000 Strain Unit. The expression of α-actin, a marker for the differentiated vascular smooth muscle cells was detected by Western blot, Real time RT-PCR and immunofluorescence. Results At 1 Hz, the expression of protein and RNA level of α-actin was higher than that of other groups. Conclusion The results demonstrate that expression of α-actin can be modulated by varying frequencies of cyclic tensile strain, which suggests varying frequency of cyclic tensile strain may take part in the modulation of phenotype transformation of vascular smooth muscle cells.
