摘  要:目的:探讨水通道蛋白-1和-5(AQP1、AQP5)在干燥综合征(Sjgrensyndrome,SS)小鼠下颌下腺中的表达及其意义。方法:选用BALB/c小鼠10只,用完全弗氏佐剂+同种鼠下颌下腺抗原诱导,使之产生在临床表现、病理损害和免疫特征上都类似于人SS的动物模型,对照组10只均注射等体积的生理盐水。用免疫组化和Western印迹方法研究SS小鼠下颌下腺AQP1、AQP5表达的变化特点。结果:SS小鼠下颌下腺组织中AQP1和AQP5的表达量均显著减少(P<0.01),主要分布在导管部位;AQP5在腺泡顶膜表达减少,而基侧膜的表达增强。结论:AQP1和AQP5表达量减少以及分布部位的异常改变,可能是SS患者口干的重要原因之一。PURPOSE: To investigate the distribution and abnormal expression of water channel proteins (AQP1 and AQP5) in the submandibular glands of mice with Sjoegren's syndrome. METHODS: A total of 10 BALB/c mice underwent injection of homologous antigen of submandibular gland tissues mixed with complete freund's adjuvant (CFA), pertussis vaccine was added to enhance the immunological reaction. Another 10 mice underwent injection of same volume of normal saline as control; 6 weeks later,the submandibular glands were examined histologically by HE staining. The sections were labeled with antibody by immunohistochemieal method and Western blot analysis. Computer-assisted image analysis system was performed to evaluate AQPI and AQP5 distribution quantitatively in the immunoreactive acini and duct of both the experimental and the control group. RESULTS: In the control group, constant and clear reaction was observed with the first antibody concentration of 1:200 for AQP5 and 1:100 for AQPI. AQPI in the acinic cell basement membrane, blood capillary and ductal basement membrane; AQP5 was strongly labeled in the acinic apical membrane and basolateral membrane,and in the secretary ducts,too;In the experimental group, Western blot found a 28kD(AQP1) and 27kD(AQP5) band in accordance with the expected molecular weight of AQPI and AQP5, and quantitative analysis of Western blot showed that the expression of AQP1 and AQP5 in the experimental group had a significant decline compared with the control group (P〈0.01); the immunoreactivity was predominantly localized to the aeinic basal membrane and ductal cells. CONCLUSION: The abnormal distribution of AQP1 and AQP5 in the aeini and ducts of the salivary gland is likely to contribute to the deficiency of fluid secretion, which is a defining feature of SS patients. The sharp decrease of AQPI and AQP5 expressed in SS patients may play an important role in leading to dry mouth.
