绍兴鸭与高邮鸭碱性磷酸酶多态性的比较

摘　　要：随机选取正常发育的30日龄绍兴母鸭和高邮母鸭各60羽,采集血液样本,置斜面上析出血清,采用聚丙烯酰胺垂直平板不连续电泳分离碱性磷酸酶。在计算机上进行条带分析并计算各泳道相对扫描灰度。结果表明:①在碱性磷酸酶电泳谱ALP-2位点上所检测的两品种鸭群中,未发现有带类型,即仅含有Alp-2°基因;②从60例30日龄绍兴鸭和高邮鸭血清样本中,在ALP-1位点上观察到F型(快带型)和S型(慢带型)2种碱性磷酸酶同功酶表现型,表现出明显的多态性;②在ALP-1位点上,高邮鸭碱性磷酸酶S型(慢带型)出现的频率显著高于绍兴鸭,但两品种鸭的基因频率(Alp和alp)无显著差异,ALP-1位点上高邮鸭基因杂合度为0．411 8,绍兴鸭为0．522 8;④另选取30、60、100、 160日龄绍兴母鸭和高邮母鸭各32羽,取血清,聚丙烯酰胺垂直平板不连续电泳,发现其各日龄表现型和基因频率无明显变化,但其扫描灰度随日龄的增长而减弱,显示碱性磷酸酶活性随日龄的增长而下降。Serum sample of 30-day Shaoxing and Gaoyou female ducks was analyzed using vertical polyacrylamide gel for the phenotypes of alkaline phosphatase （ALP） isoenzyme. The results were as follows： ① serum alkaline phosphatase isoenzyme of two ducks had no band in the locus of ALP-2, being the gene Alp-2°. ② There were both F and S phenotypes on the locus of ALP-2 in two ducks, showing polymorphism. ③ The frequency of S phenotype in Shaoxing duck was relative high than that of Gaoyou duck in the locus of ALP-1, but the frequency of gene Alp and alp showed no significant difference. The heterozygosities of Gaoyou and Shaoxing duck were 0. 411 8 and 0. 522 8. ④ Different age （30, 60, 100, 160-day） ALP scan intensities were significantly decreased both in Gaoyou and Shaoxing ducks, it showed ALP activity decreased with age.
