摘  要:采用限制性内切酶HaeⅢ、BstY Ⅰ分别对鲁西黄牛、渤海黑牛和山区水牛MHC-DRB3.2基因的Semi-Nest PCR 产物进行酶切。结果表明:3个群体均在HaeⅢ酶切位点上表现多碱基突变,山区水牛等位基因数少于前2个群体;鲁西黄牛和渤海黑牛在BstY Ⅰ酶切位点上表现多碱基突变,而山区水牛未检测到突变;X2适合性检验表明,3个群体在 HaeⅢ酶切位点上碱基突变均未达到Hardy-Weinberg平衡状态,而鲁西黄牛和渤海黑牛在BstY Ⅰ酶切位点碱基突变已达到或基本接近Hardy-Weinberg平衡。结果提示:用牛的PCR-RFLP体系可进行水牛的MHC-DRB3.2基因的研究;群体规模的急剧下降导致基因频率的失衡;山区水牛MHC-DRB3.2基因的突变率低于黄牛。The restriction polymorphisms of MHC-DRB3. 2 in 3 population of bovine were investigated by Semi-Nest PCR with enzyme of Hae Ⅲ and BstY I . The results revealed that there were extensive polymorphisms in the site of Hae Ⅲ in 3 populations, but Mountain buffalo had fewest alleles in 3 populations. Extensive mutations also existed in the site of BstY I in Luxi cattle and Bohai cattle, with homomorphism in Mountain buffalo. Statistical analysis showed that polymorphisms in the site of Hae Ⅲ did not fit with Hardy-Weinberg equilibrium in 3 populations. Polymorphisms in the site of BstY I in Luxi cattle and Bohai cattle fitted with or almost reached Hardy-Weinberg equilibrium. This study indicated the PCR-RFLP system of cattle could be applied in the research of buffalo. The decrease of population may result the genetic disequilibria. The polymorphism of MHC-DRB3. 2 in Mountain buffalo had less abundant than other two cattles.
