摘  要:目的探讨IL-1α体外诱导骨髓来源神经细胞表达神经递质的可行性。方法分离培养骨髓间充质干细胞,鉴定其表面抗原,用含30μM维甲酸的条件培养基诱导,诱导后细胞进行神经细胞特征性染色鉴定;用含25ng/ml IL-1α的培养基作用于诱导后的细胞,免疫组织化学染色鉴定神经递质的表达。结果骨髓间充质干细胞表面抗原CD29、CD44、CD105阳性,CD34、CD45、CD106阴性。诱导后细胞形态明显改变,MAP-2以及GFAP染色结果阳性,IL-1α作用后突触素染色阳性。结论IL-1α具有促进骨髓来源神经细胞表达神经递质的能力,骨髓来源神经细胞具有产生突触联系的功能。Objective The aim of this study is to investigate the feasibility of neurons derived from hMSCs to express neurotransmitter induced by interleukin-1α in vitro. Methods hMSCs were iso1αted from bone marrow, then were characterized by flow cytometry using a panel of monoclonal antibodies. Differentiation into chondrocyte-like cells was induced by cultivation of confluent cells. We characterized these cells by immunohistochemistry stain. Results Iso1αted hMSCs were positive for the markers CD105, CD29 and CD44 and negative for the markersCD34, CD45, CD106. The morphology of in- duced cells were obviously changed. Immunohistochemistry stain analysis of the confluent cells in situ showed positive for MAP2 and GFAP, also positive for synaptophysin after induced by IL-1α. Conclu- sion IL-1α has the ability to promote neurons derived from human mesenchymal stem cells to express neurotransmitter in vitro and the neurons derived from human mesenchymal stem cells have the function to emerge synaptic transmission.
