摘 要:目的探讨脑组织切片免疫组化技术中漂片法与贴片法的效果。方法建立大鼠大脑中动脉缺血模型,脑组织冰冻切片,采用兔抗BDNF(1∶500)抗体,同时运用漂片法和贴片法进行免疫组化ABC法染色。结果漂片法阳性神经元数目和灰度值与贴片法比较有显著性差异(P<0.05,P<0.01)。结论脑组织切片免疫组化技术采用漂片法效果更为可靠。Objective To explore effect of methods both bleach section and stick section of brain tissue in immunohistochemistry. Methods Adult male SD rats were created animal models of cerebral infarction in the territory of middle cerebral artery in rat. Then, constitution were prepared with frozen section and the sections were stained under the same condition using specific BDNF( 1:500)antibody by the immunohistochemistry ABC method as methods of to bleach section and to stick section were used. Results The results showed that there was a greater difference between the methods to bleach section and the methods to stick section in the number and the density of gray of BDNF positive neurons (P 〈 0.05, P 〈 0.01 ) o Conclusion It is more reliable to apply the methods to bleach section in brain tissue in technique of immunohistochemistry.
