摘  要:构建可使重组杆状病毒产生多角体的重组转移载体质粒,并将共转染产物喂昆虫幼虫,收集典型病变幼虫血淋巴进行空斑分析,发现子代病毒中约90%为重组病毒.再进行一轮空斑纯化即可获得纯净的重组病毒,较之传统的空斑纯化方法,节省了人力、物力和时间.Constuction of transfer plasmids that help recombinant baculoviruses produce polyhedra facilitates the purification of recombinant baculoviruses. Insect larvae were per os infected in this experiment with the resultant of the cotransfection of viral DNA and Plasmid DNA, and the hemolymph of diseased larvae were collected. Plaque assay showed that about 90% prgeny viruses produced in the larvae were the expected recombinant baculovirus. Another turn of plaque purification gave 100% recombinant baculoviruses. It decreases the cost and the time by this way compared to the conventional plaque assay procedure.
