摘  要:目的观察骨质疏松大鼠骨胶原结构及可溶性骨胶原超微结构的变化情况。方法实验于2003-06/12在西安交通大学医学院教育部重点实验室完成。选择SD大鼠30只,随机分2组,对照组14只,骨质疏松组16只。①骨质疏松模型复制全部大鼠经腹腔注射麻醉后打开腹腔。对照组切一小块脂肪后缝合,而骨质疏质组开腹后切除双侧卵巢后缝合。4d后进入实验,12周后麻醉处死,取骨。②模型复制成功后应用苦味酸天狼星红染色-偏振光法观察骨皮质胶原结构状况;应用骨组织胶原提取方法提取骨组织中可溶性胶原,在透射电镜下观察可溶性胶原结构。结果进入结果分析对照组大鼠12只,骨质疏松组8只,脱失动物均为取材失败或饲养问题而退出。①透射电镜下的可溶性胶原结构对照组胶原纤维横纹清晰,直径较粗。骨质疏松组横纹结构略有模糊,直径变细。②偏振光显微镜下的骨胶原排列情况对照组骨皮质胶原排列呈板层平行状,结构紧密。骨质疏松组骨皮质胶原排列不规则,结构松散,胶原甚至断裂。结论骨质疏松大鼠骨组织胶原排列紊乱,可溶性胶原超微结构模糊,直径变细,可导致骨强度下降。AIM: To observe the changes of ossein structure and dissolvable collagen ultrastructure in rats with osteoporosis. METHODS: The experiment was carried out in the Key Laboratory of Environment and Genes Related to Diseases of Ministry of Education, Xi 'an Jiaotong University between June and December 2003. Thirty SD rats were randomly divided into were divided into control group (n=14) and osteoporosis group (n=16). (1) Duplication of osteoporosis models: All the rats were anesthetized with intraperitoneal injection and then abdomen was cut open. Rats in the control group were saturated after a little fat was cut' away, and those in the osteoporosis group were saturated after bilateral ovariectomy. Four days later entered the experiment, the rats were killed under anesthesia, and then bone was removed. (2) After the successful duplication of models, picrosirius red staining and circularly polarized light were applied to observe collagen structure of cortical bone. Dissolvable collagen in bone extracted, and its structure was observed under transmission electron microscope. RESULTS: Finally 12 rats in the control group and 8 rats in the osteoporosis group were involved in the analysis of results, the other animals were excluded because of the failure of material selection and questions in feeding. (1) Structure of dissolvable collagen under transmission electron microscope: The collagen fiber stripes of control group were clear, and the diameter was great. The collagen fiber stripes of osteoperosis group were unclear and the diameter became small. (2) The arrangement of bone collagen under polarization microscope: The collagen structure of cortical bone in control group ranged regularly and tightly, and the collagen structure of cortical bone in the osteuporosis group ranged irregularly and loose. Some collagens were even broken. CONCLUSION: Osteoporosis can destroy the ossein structure and dissolvable collagen ultrastructure, narrow the diameter, and leads to the decrease
