摘 要:心肌肌钙蛋白Ⅰ具有极强的心脏特异性,可以通过检测患者血清内心肌肌钙蛋白Ⅰ的含量来诊断急性心肌梗塞。利用肌钙蛋白C和肌钙蛋白Ⅰ在钙离子存在下具有特异性结合的性质,制成了肌钙蛋白C亲和凝胶。利用肌钙蛋白C-肌钙蛋白Ⅰ亲和层析,提取和纯化了人和牛心肌肌钙蛋白Ⅰ,由100g心肌中可以提取到60mg,以后可用来制备抗人心肌肌钙蛋白Ⅰ的单克隆抗体杂交瘤细胞。Acute myocardial infarction (AMI) is suggested to be diagnosed by detecting the content of myocardial troponin Ⅰ (Tn Ⅰ ) in patient's serum, because of human cardiac specificity of myocardial Tn Ⅰ. In this paper the immobilized troponin C (TnC) affinity chromatographic gel was prepared. Relying upon the physiological properties of specific TnC-Tn Ⅰ binding in the presence of calcium human and bovine cardiac Tn Ⅰ were purified by affinity chromatography with yield of Tn Ⅰ up to 60 mg/100g cardiac muscle and the purified Tn Ⅰ was used for preparation of hybridoma cell lines which can produce monoclonal antibodies against human myocardial Tn Ⅰ for diagnosis of AMI.
