一氧化氮部分介导槲皮素的抗高血压作用

摘  要:目的:观察槲皮素的抗高血压作用及其可能机制。方法:两肾一夹复制肾性高血压大鼠模型。采用Griess反应测定NO含量,血浆及主动脉一氧化氮合酶(NOS)活性应用试剂盒测定,主动脉iNOS mRNA表达分析运用RT-PCR方法。结果:槲皮素能明显降低肾性高血压大鼠血压(174.811.9vs142.816.2mmHg,P<0.01),并能显著提高血浆NO产量及主动脉NOS活性。槲皮素能增加iNOSmRNA表达。槲皮素的这些作用可被L-NAME部分抑制。结论:槲皮素具有抗高血压作用,其机制部分涉及NO通路。Objective: Aim To investigate the antihypertensive effect and its mechanism of quercetin. Method: The Goldblatt hypertensive rat model was established by two kidneys and one clap (2K1C) . Nitric oxide (NO) content was measured with the Grieas reaction. Nitric oxide synthase (NOS) activity of plasma and aorta was determined by the guidance of assay kit. The expression of iNOS mRNA was analyzed by RT - PCR, Results: The blood pressure of hypertensive rats were decreased by quercetin ( 174. 811.9 vs 142. 816.2 mmHg, P〈 0.01) . The NO product and NOS activity in aorta were increased markedly by quercetin. The effects of quercetin were inhibited by L - NAME. The expression of iNOS mRNA was increased by quercetin in aorta. Conclusion: We conclude that quercetin has an antihypertensive effect, and its mechanism was involved in nitric oxide pathway.
