摘 要:以芦笋雄株茎尖、茎段(带一个侧芽)为外植体,研究芦笋雄株试管苗嫩茎(芽)增殖和生根的关键因子.结果表明,嫩茎(芽)增蕴较好的培养基为 MS+NAA0.1mg/L+BA0.2mg/L(或 KT0.1mg/L)+3%食用蔗糖+0.7%琼脂。用食用蔗糖代替化学纯蔗糖及自然光照代替人工照明可明显降低培养成本。品种、培养基和培养方式不同,嫩茎(芽)的增殖也不同.在生根培养中,采用二步生根法,其诱导培养基为1/2 MS+NAA0.1mg/L+IBA0.5mg/L+KT0.1mg/L+PP_(333)2mg/L+VB_i0.4mg/L+蔗糖2%+琼脂0.6%,时间9~11天;生根培养基为1/2 MS+KT0.1mg/L+PP_(333)2mg/L+VB_10.4mg/L+AC0.005%+蔗糖2%+琼脂0.6%,时间19~21天.二步生根法的生根率达80%以上,显著高于一步生根法;且根系发达,素质好,培养时间仅为一步生根法的一半。The apical mcristem or stem with one lateral bud was used as explant to study the key factors to the multiplication and rooting of asparagus in vitro.The results showed that the culture medium suita- ble for multiplication was MS+NAA 0.1mg/L+BA 0.2mg/L(or KT 0.1mg/L)+diet sugar 3%+Agra 0.7%.By using diet sugar and natural light instead of pure sugar and artificial light could greatly reduce the cost.The multiplication ability of asparagus differed from cultivars,composision of culture media and methods of cultivation.A two-step rooting method was developed,which is discribted as following:the explants were cultured in the pre-rooting culture medium of 1/2 MS+NAA 0.1mg/L+IBA 0.5mg/L +KT 0.1 mg/ L+PP_(333)2mg/L+VB_1 0.4mg/L+sugar 2%+Agar 0.6% for 9-11 days,then transfered to the rooting culture medium of 1/2 MS+KT 0.1mg/L+PP_(333)2mg/L+VB_1 0.4mg/L+AC0.005% +Sugar 2%+Agar 0.6% for 19-21 days.Results indicated that the percentage of rooting explants was above 80%,in the two-step rooting method,higher than the one-step,with a better root systems.The cultivation period could be shortened to about half of that in one-step method.
