摘 要:目的:研究海藻繁枝蜈蚣藻抗单纯疱疹病毒Ⅰ型活性。方法:采用空斑减数实验测定5种不同提取方法所得蜈蚣藻粗提物抗HSV-1活性,计算空斑抑制率和ED50,并从药物对细胞的保护、对病毒增殖的影响及对病毒感染细胞的综合作用3个方面初探繁枝蜈蚣藻抗单纯疱疹病毒活性机理。结果:蜈蚣藻多糖能明显抑制HSV-1的致病变作用,ED50依次为8.72、12.16、8.93、8.49和805 982.62μg/m l,药物在对细胞的保护及对病毒感染细胞的综合作用中均表现出明显活性。结论:繁枝蜈蚣藻有显著的抗HSV-1活性,且初步推测蜈蚣藻多糖抗HSV-1活性是作用在病毒和受体结合、侵入Vero细胞阶段。Objective:To investigate the activities of anti - Herpes simplex Ⅰ virus of amyloses axtracted from Grateloupiafilicina via five kinds of extraction methods. Methods:HSV - 1 was inoculated on Vero cells, Amylase axtracted from Grateloupiafilicina was added to the medium in various concentrations, after 72 hours incubation, plaques were counted and the 50% effective dose (ED50) was calculated in plaque reduction assay. Initial studies on antivirus mechanism in three aspects: the drug protective action, the drug influence on virus proliferation and the drug synthesis action on virus were studied. Results:Amyloses had conspicuous anti - Herpes simplex Ⅰ virus activities, their ED50 were 8.72, 12. 16, 8.93, 8.49 and 805 982. 62μg/ml, respectively. Amyloses had antivirus activities in the drug protective action and the drug synthesis action on virus, however, had not in the drug influence on virus proliferation, Conclusion: Gratelottpia filicina has conspicuous anti -Herpes simplex Ⅰ virus activity, and the activity happened in the stage of HSV - 1 binding, adsorpting with Vero cell.
