摘 要:目的:探讨双黄连粉针剂在体外是否有抗HIV作用及其可能的作用机制。方法:采用荧光染料Calcien AM标记HIV感染的慢性H9细胞(H9/HIV-1ⅢB)与不同稀释浓度的双黄连粉针剂作用后,与靶细胞MT-2细胞混合培养,观察MT-2和H9/HIV-1ⅢB融合细胞的变化;MTT法检测双黄连对HIV感染细胞的保护作用;HIV-1 P24抗原试剂盒检测药物作用后的细胞培养上清P24抗原含量的变化。结果:双黄连粉针剂能够抑制MT2和H9/HIV-1ⅢB融合,能够保护HIV感染细胞的死亡,减少HIV-1 P24抗原的表达。结论:双黄连粉针剂在体外具有抗HIV-1的作用。Objective: To investigate the inhibitory effects of Shuanghuanglian (SHL) injection on human immunodeficiency virus type 1 (HIV- 1) infection in vitro and the possible mechanisms. Methods: Calcein- AM stained H9/HIV- 1Ⅲ B cells were treated with serially diluted SIlL injection, then mixed with target ceils MT - 2, HIV - 1 mediated cell fusion was observed under fluorescent microscopy. The protecting effect of SHL on HIV - 1 - infected cells was determined by MTT. Viral replication was evaluated by mea suring the levels of p24 antigen in culture supematants by ELISA, Results: SHL injection could inhibit fusion of MT- 2 and H9/HIV 1Ⅲ B ceils, could protect HIV linfected ceils from death, and decrease p24 antigen release. Conclusion: SHL injection can reduce HIV- 1 infection in vitro.
