摘  要:骨形态发生蛋白15(bone morphogenetic prote in 15,BMP15)基因为控制Belc lare和Cambridge绵羊高繁殖力的主效基因。采用PCR-RFLP和PCR-SSCP技术检测BMP15基因中与绵羊高繁殖力相关的两个突变位点在高繁殖力山羊品种济宁青山羊(130只母羊、20只公羊)以及低繁殖力山羊品种(波尔山羊30只母羊、文登奶山羊30只母羊、辽宁绒山羊30只母羊、内蒙古绒山羊50只母羊、北京本地山羊30只母羊)中的单核苷酸多态性,同时研究这两个突变位点对济宁青山羊高繁殖力的影响。结果表明,在6个山羊品种中都没有检测到BMP15的B2突变(C→T);检测到的BMP15的B4突变(G→T)在6个山羊品种中均呈现杂合子状态。这表明BMP15基因中影响Belc lare和Cambridge绵羊高繁殖力的突变位点对济宁青山羊的高繁殖力没有显著影响。Bone morphogenetic protein 15 (BMP15) gene which controls the fecundity of Belclare ewes and Cambridge ewes was studied as a candidate gene on the prolificacy of Jining Grey goats. Single nucleotide polymorphism ofBMP15 gene was detected in both high fecundity goat breed( 130 does and 20 bucks of Jining Grey goat) and low fecundity goat breed (30 Boer does,30 Wendeng dairy does,30 Liaoning Cashmere does, 50 Inner Mongolia Cashmere does,30 Beijing native does) by PCR-RFLP and PCR-SSCP. The results showed that the B2 mutation (C→T) of BMPI5 gene was not detected in six goat breeds. The B4 mutation (G→T) of BMP15 gene was detected, the B4 mutations were all heterozygous in six goat breeds. These results indicated that these mutations had significant effects on fecundity of Belclare ewes and Cambridge ewes had no Significant effect on prolificacy of Jining Grey goats.
