温肾补阳法对肾阳虚变应性鼻炎大鼠模型血清Th_1/Th_2细胞因子表达的影响

摘　　要：目的研究肾阳虚因素对Th1/Th2细胞因子平衡的影响,以及温补肾阳法对Th1/Th2细胞因子平衡调控作用,为变应性鼻炎的中医病因学说和治疗方法提供科学依据。方法在腺嘌呤致肾阳虚病理模型的基础上,用卵清蛋白为过敏原对大鼠致敏,建立肾阳虚变应性鼻炎大鼠模型。以温补肾阳代表方剂金匮肾气丸灌胃,观察其行为学差异,进行鼻腔分泌物涂片及鼻腔黏膜组织学检查;用酶联免疫吸附法(ELISA)对血清中Th1细胞因子IFN-γ、IL-2和Th2细胞因子IL-4、IL-5进行检测。结果变应性鼻炎组和肾阳虚变应性鼻炎组行为学积分明显高于温补肾阳组和正常对照组,差异有显著性(P<0.05);温补肾阳组鼻黏膜炎性反应明显轻于变应性鼻炎组、肾阳虚变应性鼻炎组;温补肾阳组IFN-γ、IL-2水平明显高于变应性鼻炎组和肾阳虚变应性鼻炎组,差异有显著性(P<0.05),与正常组比较无显著性差异;温补肾阳组IL-4、IL-5表达水平明显低于变应性鼻炎组和肾阳虚变应性鼻炎组,差异有显著性意义(P<0.05),与正常组比较无显著性差异。结论温肾补阳法能调节Th1和Th2细胞因子的表达,纠正失衡的Th1/Th2的细胞因子网,因而对变应性鼻炎产生治疗作用。Objective To find out the relationship between kidney - yang - deficiency syndrome and the Th1/Th2 expression, and to investigate the effect of kidney - yang warming therap yon the Th1/Th2 expression. Methods Kidney - yang - deficiency rat models were firstly induced by adenine and then were sensitizated by inhalation of ovalbumin to establish allergic rhinitis （AR） models . The treatment group was given Jinkui Sheruti pill. After treatment , the changes of behavior, secretion of nasal cavity and nasal mucosa were observed. The levels of Th1 cytokines IFN-γ, IL- 2 and Th2 cytokines IL- 4 , IL- 5 in serum were determined by ELISA. Results The behavioral score in AR group and kidney- yang- deficiency AR group was higher than that in the treatment group and normal control group （ P 〈 0. 05） . Inflammatory reaction in nasal mucosa of the treatment group was remarkably milder than that in AR group and kidney - yang - deficiency AR group. The mean levels of serum IFN-γ, IL - 2 in the treatment group were significantly higher but those of IL - 4 , IL - 5 levels much lower（ P 〈 0. 05） than those in AR group and kidney - yang - deficiency AR group, the difference of the above indexes was insignificant between the treatment group and the normal control group. Conclusion The kidney - yang - defiency syndrome has a relation with Th1 / Th2 imbalance. The kidney - yang warming therapy is effective for kidney - yang - deficiency allergic rhinitis, and its mechanism may be related to the regulation of the expression of Th1 and Th2 cytokines and correction of the imbalance of Th1/ Th2 cytokines network.
