摘 要:目的建立藏药独一味及其制剂中总环烯醚萜苷的含量测定方法。方法采用分光光度法,以8-乙酰氧基山栀苷甲酯(8-O-acetylshanzhisidemethylester)为对照品,75%乙醇为反应溶剂,加入50%HCl,80℃水解20min,以对二甲氨基苯甲醛为显色剂,在620nm处测定其含量。结果8-乙酰氧基山栀苷甲酯显色化合物稳定,在5.1~42.0μg/mL范围内有良好的线性关系(r=0.999);平均回收率为102.16%,RSD为3.92%。结论该实验建立了对二甲氨基苯甲醛比色法测定独一味及其制剂中总环烯醚萜苷含量的方法,该方法简便、可靠,可作为独一味及其制剂中总环烯醚萜苷的质量控制方法。Objective To establish a method for the determination of the total iridoid glycoside in Tibetan herbal medicine of Lamiophlomis rotata and its preparation. Methods The samples were hydrolyzed by 50 % HCl at 80 ℃ for 20 min. Spectrophotometry was established to determine the total iridoid glycoside . In this method, 8- O- acetyl shanzhiside methylester was used as the reference substance, 75 % ethanol as reaction solvent, and paradimethylaminobenzal dehyde as color - developing agent. The maximal absorption wavelength was 620 nm. Results Coloration compound of 8 - O - acetyl shanzhiside methylester was stable. There was a good linearity in the range of 5. 1 - 42μg/mL ( r = 0. 999) , The average recovery was 102.9 % and RSD was 6. 40 %. Conclusion With paradimethylaminobenzal dehyde as the color developing reagent, spectrophotometry is established firstly for the determination of total iridoid glycoside in Tibetan herbal medicine of Lamiophlomis rotata and its preparation. This method is effective for the quality control of the total iridoid glycoside in herb Lamiophlomis rotata and its preparation.
