抗角蛋白自身抗体对培养角朊细胞产生白细胞介素8的影响

摘　　要：为探讨抗角蛋白自身抗体（AKantoAb）对角朊细胞免疫功能的影响，实验以IL-1β刺激无血清培养角朊细胞及AKautoAb作用后的角朊细胞，用夹心ELISA法检测培养上清中IL－8的产量。结果表明IL-1β可刺激角朊细胞产生IL－8，并对IL－1β有浓度依赖性，AKautoAb对角朊细胞产生IL－8有明显抑制作用。提示AKautoAb在炎性皮损角朊细胞中的沉着，可能存在有益的调节作用。This study,one part of systematical studies of antikeratin auto antibody (AK auto Ab), was aimed at investigating the effects of AK auto Ab on the immunological capabilities of keratinocytes. Keratinocytes were cultured in serum free keratinocyte medium and cultured keratinocytes pretreated by Ak auto Ab were stimulated by interleukin-1β. The quantity of interleukin-8 in extract was measured by enzyme-linked immunosorbent assay (ELISA). The results showed that IL -1β induced the production of IL8 in a dose dependent manner; 800 u/ml and higher concentrations IL-1β could increase the production of IL-8 (P<0. 01 ); Ak auto Ab inhibited keratinocytes IL-8 production stimulated by IL-1β (P < 0.01 ). It was suggested that in inflammatory dermatitis Ak auto Ab deposited in keratinocytes in lesions may regulate the inflammatory response through inhibiting the IL-8 production by keratinocytes.
