摘 要:目的了解皖籍汉族正常人群中MICA(majorhistocompatibilitycomplexclasschain-relatedgeneA)等位基因的分布,为进一步研究MICA关联疾病的发病机制奠定基础。方法采用聚合酶链反应-序列特异性寡核苷酸探针杂交(polymerasechainreactionandsequence-specificoligonucleotideprobing,PCR-SSOP)技术对112例正常人群进行MICA基因外显子2、3、4的多态性分析。结果本研究人群的MICA基因多态性以MICA*008等位基因频率最高,达30.36%,其次为*010(16.96%)、*002(13.84%)和*009(12.05%)。单倍型分析显示,MICA基因与多个HLA-B位点间存在连锁不平衡现象,有统计学意义的有MICA*008-B40、B07、B13,MICA*010-B46、B15,MICA*002-B35、B15和MICA*009-B52。结论皖籍汉族正常人群的MICA等位基因分布具有其基本特点,MICA基因与HLA-B位点间存在广泛连锁不平衡。Objective In order to know genetic polymorphism in the exons 2 to 4 of the MICA gene in Han national ity population from Anhui province and play a basis on the study of pathogeny of diseases associated with MICA allele. Method DNA samples from 112 random healthy individuals,unrelated Han nationality population were genotyped by PCR-SSOP to analyse the frequencies of MICA alleles. Results MICA*008 was dominant in MICA allele ,accounted for 69.8% ,the others were MICA* 010(16.96% )〉* 002(13. 84%)〉* 009(12.05%) respectively. The haplotype analysis revealed that linkage disequilibrium were lied in MICA *008-B40 ,B07, B13, MICA*010-B46, B15, MICA*002-B35, B15 and MICA*009-B52. Conclusion The distribution of MICA alleles has basic characteristics in Han nationality population from Anhui province.
