反相高效液相色谱外标法测定苯巴比妥与卡马西平的血药浓度

摘　　要：目的:建立反相高效液相色谱外标法,测定苯巴比妥与卡马西平的血药浓度。方法:选用醋酸乙酯为萃取溶剂,采用外标法进行含量测定。色谱柱为SymmetryC18柱(4.6mm×150mm,5.0μm),流动相为甲醇-水(50∶50),流速为1mL/min,检测波长为254nm。结果:苯巴比妥与卡马西平线性范围分别为1～100μg/mL和1～50μg/mL,平均回收率分别为99.95%和99.65%,日内、日间RSD均小于5%,标准曲线相关性良好。结论:该法简便、经济,结果准确,适合临床应用。Objective： To establish a simple method of detecting phenobarbital and carbamazepine in serum by external reference method of RP- HPLC. Methods： Ethyl acetate as extract solvent, drug content was detected by external reference method with analytical column Symmetry C18 column（4.6 mm× 150 mm, 5.0μm）, mobile phase methanol-water（50 ： 50）, flow rate 1 mL/min and detection wavelength 254 nm. Results： By this method, the linear ranges of phenobarbital（PB） and carbamazepine（CBZ） were 1- 100 and 1 -50 μg/mL respectively. The average recovery rate of PB and CBZ were 99.95% and 99.65% respectively. The relative standard deviations（RSD）of within day and inter-day were less than 5%. The calibration curve presented good linearity. Conclusion： It proves a simple, economical, accurate and sensitive method and suitable for clinical application.
