摘  要:目的观察正常大鼠海马NMDA受体2B亚单位(NR2B)蛋白质及其mRNA的基础性表达特征及两者在海马不同区域的表达关系。方法正常SD大鼠海马,灌注取脑,连续冰冻切片,ABC免疫组织化学方法染色和原位杂交组织化学染色,光镜下观察NR2B蛋白质及其mRNA的表达。结果NR2B蛋白质在正常大鼠海马CA1区、CA3区及齿状回的免疫反应强度存在明显差异,CA1区最强,CA3区次之,齿状回着色较弱。NR2B mRNA在正常大鼠海马CA1区、CA3区及齿状回的原位杂交组织化学反应强度与NR2B蛋白质免疫反应强度存在平行变化,也是CA1区最强,CA3区次之,齿状回着色较弱。结论NR2B蛋白质及其mRNA在正常大鼠海马CA1区、CA3区及齿状回的表达强度不同,并存在平行表达差异关系,这种基础状态的分布特征可能与海马缺血易损性和耐受性相关。Objective To study the normal morphological distribution of immunoreactive protein and mRNA of NMDA receptor 2B subunit (NR2B) in rat hippocampus to investigate the relation between the protein and the mRNA. Methods After transcardiac perfusion with 4% paraformaldehyde solution, the brain was removed to cut serial cryo - sections, which were processed by ABC immunohistochemistry and in situ hybridization (ISH) to study the expression of NR2B protein and its mRNA respectively. Results NR2B and NR2B mRNA were widely distributed in all normal hippocampal subfields. The immunoreactivity of NR2B was stronger in the CA1 than in the CA3 subfield, but was rather weak in the dentate gyrus. The expression of NR2B mRNA in ISH was parallel to the immunoreactivity of NR2B. Conclusion It seems that the characters of the anatomical distribution of NR2B and NR2B mRNA might be relevant to the vulnerability of the CA1 subfield to ischemic injury.
